Establishment of early brain injury model after spontaneous subarachnoid hemorrhage in rabbits

摘  要:目的建立一种蛛网膜下腔出血(SAH)后早期脑损伤动物模型。方法选用新西兰大耳白兔60只,随机分为实验组36只,对照组18只,空白组6只。实验组采用枕大池穿刺一次注入自体动脉血法建立SAH模型,对照组注入平衡液,空白组仅行枕大池穿刺。在造模后4、8、12、24、48、72 h,观察比较各组动物神经功能改变和脑组织形态学变化。结果肉眼观察实验组造模后见蛛网膜下腔大量血凝块或血液,光镜和电镜下见蛛网膜下腔大量红细胞,神经细胞水肿、变性;对照组和空白组无异常。实验组注血后出现明显的神经行为学症状;与对照组比较,实验组神经功能评分在8~72 h明显升高(P〈0.01);与空白组比较,实验组神经功能评分在各时间点均明显升高(P〈0.01)。结论枕大池一次注入自体动脉血法建立自发性SAH模型简便、可重复,适宜于研究SAH后早期脑损伤。Objective To establish a new animal model of early brain injury after symptomatic subarachnoid hemorrhage(SAH) in rabbits.Methods Sixty rabbits were divided randomly into 3 groups: 36 rabbits in experimental group,18 in control group and 6 in blank group.SAH models were set up through the method for cisternal puncturing and one-time injecting autologous arterial blood in experimental group,while injection of balanced solution was performed in control group and only cisternal puncture in blank group.The neural function and histomorphology of the brain tissue were observed and compared 4,8,12,24,48 and 72 h after modeling.Results A lot of blood clots or blood were found by naked eyes,and plenty of large red blood cells,edematous and denatured nerve cells under light or electron microscope in experimental group after SAH.However,no abnormality was observed both in control group and blank group.Significant neuroethological symptoms occurred after SAH in experimental group.Compared with the control group,the score of neurological function rose significantly during 8 to 72 h after SAH in the experimental group(P0.01).Compared with the blank group,the score of neurological function significantly increased in any time point after SAH in the experimental group(P0.01).Conclusions Cisternal puncture and one-time injection of autologous arterial blood is a simple and repeatable method for SAH model,and suitable for research of early brain injury.
