摘 要:采用梯度压力式驯化法,从河南某印染厂的活性污泥中富集筛选一组活性红BF-3BN高效脱色菌群AR1。经菌株分离和16S rDNA测序分析,确定菌群内微生物分别归属Clostridium sp.、Pseudomonas sp.和Shewanella sp.等3个属。染料脱色实验结果表明,在活性红BF-3BN起始浓度为200 mg/L的脱色培养基中,菌群AR1脱色的适宜pH 5~10,温度25~35℃,时间24 h,能达到95%以上的最大脱色率。该菌群对高浓度活性红BF-3BN有较好的脱色效果,当活性红BF-3BN浓度低于500 mg/L时,脱色率高于90%,即使在含有600 mg/L活性红BF-3BN的脱色培养基中,脱色率仍然能达到80%以上。A bacterial flora AR1 capable to decoloring reactive red BF-3BN effectively was screened from the activated sludge of dyeing wastewater treatment in Henan Province by the dye gradient domestication method.In order to determine the composition of the flora,three isolates were detected by plate isolation.The phylogenetic analysis on the basis of 16S rRNA gene sequence showed that the closest relative in the flora AR1 belonged to Clostridium sp.,Pseudomonas sp.and Shewanella sp.three genera.The results of the dye decolorization experiments suggested that the flora AR1 could give a maximal decolorization rate of 95% for reactive red BF-3BN of 200 mg/L under the optimal culture conditions: inoculation volume of 10%(V/V),pH of 5 to 10,temperature of 25 to 30 ℃ and culture time 24 h.The decolorization rate of BF-3BN by AR1 was higher than 90% when the initial concentration of BF-3BN was lower than 500 mg/L,and it could also maintain 80% even the initial concentration of BF-3BN was 600 mg/L.
