摘 要:目的探讨索拉非尼联合热疗对肝癌HepG-2细胞的抑制作用。方法实验细胞分为4组:对照组(正常培养的HepG-2细胞)、索拉非尼组(索拉非尼药物浓度为12μmol/L)、热疗组(加热温度为43℃,加热时间为2 h)及索拉非尼联合热疗组。以MTT法测定HepG-2细胞的增殖抑制情况,用流式细胞仪检测细胞凋亡情况。结果①索拉非尼组、热疗组、索拉非尼联合热疗组细胞增殖均明显受到抑制,与对照组比较差异有统计学意义(P<0.01)。以索拉非尼联合热疗组抑制作用最为显著,与索拉非尼组和热疗组比较差异有统计学意义(P<0.01),索拉非尼和热疗两种处理因素对细胞增殖抑制存在交互作用(F=129.183,P<0.01)。②与对照组比较,索拉非尼组、热疗组及索拉非尼联合热疗组细胞的早期凋亡率均明显增加,差异有统计学意义(P<0.01);与索拉非尼组和热疗组比较,索拉非尼联合热疗组早期凋亡更加明显(P<0.01),索拉非尼和热疗两种处理因素对细胞凋亡诱导存在交互作用(Annexin V-EGFP染色,F=197.630,P<0.01;PI染色,F=643.210,P<0.01)。结论索拉非尼和热疗对肝癌HepG-2细胞均有增殖抑制作用及促凋亡作用,索拉非尼联合热疗具有协同作用。Objective To investigate the inhibitory effect of Sorafenib combined with thermotherapy on hepatocellular carcinoma HepG-2 cells.Methods The experiment cells were divided into 4 groups: control group(normal cultured HepG-2 cells),Sorafenib group(concentration of Sorafenib was 12 μmol/L),thermotherapy group(heating temperature was 43℃,and heating time was 2 h) and Sorafenib combined with thermotherapy group.The proliferation inhibition in HepG-2 cells was determined by MTT assay and the apoptosis was detected by flow cytometry(FCM).Results ①Compared with that of control group,the proliferation was inhibited in Sorafenib group,thermotherapy group and Sorafenib combined with thermotherapy group(P0.01),especially in Sorafenib combined with thermotherapy group,and the differences between Sorafenib combined with thermotherapy group and Sorafenib group,and Sorafenib combined with thermotherapy group and thermotherapy group were significant(P0.01).Sorafenib and thermotherapy had interactive effect in cytostasis(F=129.183,P0.01).②Compared with that in control group,the early apoptosis rate in the other 3 groups were increased obviously,and the differences were significant(P0.01);Compared with that in Sorafenib group and thermotherapy group,the early apoptosis was significant in Sorafenib combined with thermotherapy group(P0.01).Sorafenib and thermotherapy had interactive effect in inducing apoptosis(Annexin V-EGFP staining,F=197.630,P0.01;PI staining,F=643.210,P0.01).Conclusion Both Sorafenib and thermotherapy can inhibit the proliferation and induce the apoptosis of hepatocellular carcinoma HepG-2 cells,and Sorafenib combined with thermotherapy has some synergistic effect.
