慢病毒介导siRNA干扰Sez6对小鼠小脑浦肯野细胞发育的影响

摘  要:目的通过Sez6 siRNA慢病毒感染小鼠小脑组织切片细胞,干扰Sez6基因表达后,观察浦肯野细胞生长发育变化。方法使用Sez6 siRNA慢病毒感染体外培养的刚出生(P0)的小鼠小脑组织切片细胞后;免疫组化标识浦肯野细胞,观察细胞树突发育。结果 Sez6基因表达下调明显,浦肯野细胞树突分支数量减少(P<0.05),发育延迟;浦肯野细胞胞体排列不整齐。结论 Sez6的表达与浦肯野细胞树突发育相关,还可能跟浦肯野细胞迁移排列相关。Objective To observe the development of Purkinje cells in mouse cerebellar tissue slices after the interference of Sez6 gene expression by Sez6 siRNA mediated with lentivirus in vitro. Methods The mouse cerebellar tissue slice were infected by Sez6 siRNA lentivirus in vitro. The development of dendrites in Purkinje cells were examined by immunohistochemistry. Results The expression of Sez6 in Purkinje cells was down-regulated significantly after infected with Sez6 siRNA lentivirus. The development of dendrites in Purkinje cells was delayed, and the quantity of dendrites reduced ( P 〈 0.05 ). The arrangement of the Purkinje cell bodies was irregular. Conclusion The gene expression of Sez6 might be associated with the development of dendrites in Purkinje cells, and also with the migration and arrangement of Purkinje cell bodies in mouse cerebellum.
