摘 要:利用电喷雾多级串联质谱法研究了中药药对共煎液中双酯型、单酯型及脂型生物碱的生物转化过程,建立了乌头类生物碱生物转化前后的质谱半定量方法,分析了大鼠肠内菌群作用下制川乌分别与浙贝母、法半夏和白蔹配伍后共煎液的生物转化过程中乌头类生物碱的含量变化.结果表明,在配伍后的共煎液中,浙贝母可增加双酯型生物碱含量,法半夏可降低双酯型生物碱含量,而白蔹对双酯型生物碱含量影响不大;大鼠肠内菌群能够进一步降低双酯型生物碱的毒性并增加脂型生物碱的含量.本文通过分析中药药对中乌头类生物碱含量的改变说明了中药配伍及生物转化机理.The biotransformation of diester-diterpenoid alkaloid,monoester-diterpenoid alkaloid and lipo-alkaloid in the decoctions of Chinese medicine pairs by rat intestinal flora was studied using electrospray ionization tandem mass spectrometry.The semiquantitative method of mass spectrometry was established for the analysis of aconitum alkaloids before and after biotransformation.The content changes of aconitum alkaloids caused by rat intestinal flora was analyzed in the biotransformation process of the decoctions of Radix Aconiti Preparata with Bulbus Fritillariae Thunbergii,Rhizoma Pinelliae Preparatum and Ampelopsis Japonica.The results showed that after the combination Bulbus Fritillariae Thunbergii increased the content of diester-diterpenoid alkaloids,and Rhizoma Pinelliae Preparatum reduced the content of diester-diterpenoid alkaloids,and Ampelopsis Japonica almost has no effect for the content of diester-diterpenoid alkaloids in decoction;the rat intestinal flora can more reduce the toxicity of diester-diterpenoid alkaloids and increase the content of lipo-alkaloids.The mechanism of combination and biotransformation of traditional Chinese medicine was proposed by the analysis of the content changes of aconitum alkaloids in Chinese medicine pairs.
