摘 要:目的:观察RNA干扰(RNA interference)对人胆囊癌GBC-SD细胞株中人端粒酶逆转录酶(human telomerase reverse transcriptase,hTERT)表达的抑制作用。方法:设计靶向hTERT基因的微小RNA(miRNA),构建4个重组表达质粒并转染人胆囊癌GBC-SD细胞株。48h后用RT-PCR和Western blot方法检测hTERT的mRNA和蛋白表达情况。结果:与对照组相比,靶向hTERT的4个重组质粒转染组的hTERT mRNA水平和蛋白表达量均明显降低(P<0.01)。结论:靶向hTERT基因的特异性miRNA表达载体能有效地下调GBC-SD细胞中hTERT mRNA及蛋白表达水平。Objective To evaluate the inhibitory effect of RNA interference on the expression of human telomerase reverse transcriptase (hTERT) mRNA and protein in human gallbladder cancer GBC-SD cell lines. Methods After miRNA targeting hTERT gene was designed, 4 recombinant plasmid vectors were constructed and transfeeted into human gallbladder cancer GBC-SD cells. After 48 hours, mRNA and protein levels of hTERT were determined by RT-PCR and western blot techniques, respectively. Results As compared with control group, the expressions of hTERT mRNA and protein were both significantly decreased after treated by 4 recombinant plasmids hTERT-targeted miRNA-1,-2,-3 and-4 (P 〈 0.01 ). Conclusion The miRNA expression vector targeting hTERT gene could effectively down-regulate the expression of hTERT mRNA and protein in GBC-SD cells.
