摘 要:目的:探讨纳米脂质体槲皮素(nLQ)和8-Br-cAMP(Br)对人视网膜母细胞瘤Rb44细胞凋亡和p53、p21waf1mRNA表达及Bcl-2、Bax和Caspase-3蛋白表达的影响。方法:Rb44细胞分为4组:nLQ组给予终浓度为40μmol/LnLQ;Br组给予终浓度为2×10-5mol/LBr,联合用药组给予同上浓度的nLQ和Br,对照组不给药。采用TUNEL法检测各组细胞的凋亡率。采用原位杂交技术检测p53、p21waf1mRNA的表达,免疫细胞化学方法检测Bcl-2、Bax及Caspase-3蛋白的表达。结果:与对照组相比,单用nLQ或Br均可增高凋亡率,上调p53和p21waf1mRNA的表达及Bax和Caspase-3蛋白的表达,下调Bcl-2蛋白的表达(P<0.001);nLQ及Br联用呈现协同效应(P<0.001)。结论:nLQ和8-Br-cAMP可能通过上调p53和p21waf1基因的表达,诱导线粒体途径和Caspase-3的最终共同凋亡途径而诱导Rb44细胞趋向凋亡,且两药联用呈现协同效应。Aim:To explore the synergistic effects of the nanoliposomal quercetin(nLQ) and 8-Br-cAMP(Br) on apoptosis and expressions of p53 mRNA,p21waf1 mRNA,Bcl-2,Bax and Caspase-3 in Rb44 cells.Methods:Rb44 cells were allocated into 4 groups:nLQ group (given 40 μmol/L nLQ),Br group (given 2×10-5 mol/L Br),combined group(given the 2 drugs),and control group (not treated).The apoptosis was detected by TUNEL.The mRNA expressions of p53 and p21waf1 were examined by in situ hybridization.The expressions of Bcl-2,Bax and Caspase-3 were detected by immunocytochemistry.Results:Compared with control group,the apoptotic rate increased,the mRNA expressions of p53 and p21waf1 was upregulated,the expression of Bcl-2 was down-regulated,and those of Bax and Caspase-3 were promoted in nLQ and Br groups(P0.001).Moreover,nLQ and Br combined use had synergistic effects (P0.001).Conclusion:nLQ and Br could induce apoptosis with synergistic effects through mitochondria and final common apoptotic pathway.
