摘 要:目的探讨KK-AY糖尿病小鼠视网膜中细胞间黏附分子-1(ICAM-1)和血管内皮细胞生长因子(VEGF)表达情况。方法实验共分为4组,每组10只小鼠。对照组为12周的C57BL/6小鼠;糖尿病模型组为KK-AY小鼠,分别为12周、16周、20周组。测量各实验组体重、血糖,并应用逆转录聚合酶链式反应(RT-PCR)和蛋白免疫印迹(Western blot)方法检测视网膜中ICAM-1和VEGF mRNA和蛋白的表达情况。结果糖尿病模型组12周时视网膜ICAM-1和VEGF表达均出现上升,16周时继续增加,20周时分别到达其最高点,其中16周、20周组与对照组相比差异均有统计学意义(P<0.05)。结论 ICAM-1和VEGF可能参与KK-AY小鼠糖尿病视网膜病变的发生。Objective To study the expression of intercellular adhesion molecule-1(ICAM-1) and vascular endothelial growth factor(VEGF) in retina of KK-AY mice.Methods Experimental mice were divided into 4 groups: C57BL/6(control group) and KK-AY diabetic mice groups(12-week,16-week,20-week group),10 mice in each group.The body weight and blood glucose of mice were measured,meanwhile,the expression of ICAM-1 and VEGF in each group was observed using reverse transcription-polymerase chain reaction(RT-PCR) and western blot analysis.Results The expression of ICAM-1 and VEGF increased in 12-week KK-AY mouse retina,with higher expression in 16-week group,and had the highest expression in 20-week group.The difference between the control group and the 16-week,20-week groups was significant(P0.05).Conclusion ICAM-1 and VEGF may participate in the development of diabetic retinopathy.
