摘 要:目的检测miR-141,miR-16,miR-103,miR-574,miR-34b等五种前列腺癌相关的miRNA在前列腺癌患者血清中的含量,评价其在前列腺癌诊断中的价值。方法收集临床血清样本,将其分为前列腺癌、前列腺增生,PSA升高等三组,提取血清中miRNA。应用荧光定量PCR方法检测相应miRNA的含量,用GraphPadPrism软件进行统计学分析。结果miRNA-103和miR-34b在前列腺癌患者、前列腺增生患者和PSA增高人群血清中的含量普遍较正常人增高,其中miR—NA-34b平均增高2.2×10^4倍,在所有PSA升高的样本中均增高。其余3种miRNA的水平相对于正常人未呈现显著差异。结论miR-34b的诊断价值与PSA相当,是一个潜在的前列腺癌诊断性血清标志物。Objective To detect the serum level of miRNA-141,miRNA-16,miRNA-103,miRNA-574 and miRNA-34b in prostate cancer patients. Then evaluate the value of the miRNA markers in prostate cancer diagnosis. Methods Total RNA including miRNA was extracted from serum samples of normal people, benign prostatic hyperplasia (BPH) and prostate cancer (PCa) patients. The serum levels of the 5miRNA were detected using fluorescence real-time PCR. And the association between miRNA markers and cancer development was analyzed using GraphPad Prism software. Results The serum level of miRNA-103 and miRNA-34b were significantly up-regulated in BPH and PCa patients. Especially miRNA- 34b in PCa increased average 2.2 × 10^4 folds than in normal people,and correlated with high PSA. All the other 4 miRNA did not change a lot in BPH and PCa patients comparing with normal people. Conclusion miRNA-34b is a potential diagnostic serum marker of prostate cancer,might be equal to PSA.
