摘 要:对黑颈鹤病原菌进行快速鉴定及毒力因子分析。采用16S rRNA序列分析法鉴定黑颈鹤病原菌,其毒力基因分别用大肠杆菌的irp2、papC、iucD、tsh、iss毒力基因的特异性引物进行PCR检测,并将扩增出来的irp2基因序列与Genbank^(TM)相应序列进行同源性分析。分离菌株鉴定为大肠埃希氏菌,鉴定结果与生化检测结果一致,分离菌携带irp2毒力基因,这从毒力试验得到证实,其序列与与Genbank^(TM)上发表的HPI irp2基因序列同源性高达98%以上。采用16S rRNA序列分析法可以对黑颈鹤大肠埃希氏菌感染进行快速鉴定,黑颈鹤源性大肠杆菌携带有irp2毒力基因。We analyzed pathogens isolated from black-necked crane.We identified pathogens by 16S rRNA sequence analysis. Specific primers of irp2,papC,iucD,tsh,and iss virulence genes were used to detect virulence genes of the pathogen using PCR.Sequence homology was compared between the amplified irp2 gene sequence and irp2 sequences published on Genbank^(TM). The isolated pathogen was identified as Escherichia coli,identical to the outcome of a biochemical test.The pathogen carried the irp2 gene,which was also confirmed from the toxicity test.The homology rate was above 98%in comparison with the HPI irp2 sequences published on GenBank^(TM).We conclude that the 16S rRNA gene sequence analysis can be used for rapid identification of E.coli,and that E.coli isolated from black-necked crane Carries the irp2 virulence gene.
