CEA-rv诱导特异性CIK细胞对Lovo细胞株杀伤作用的研究

摘　　要：目的探讨癌胚抗原重组基因痘苗(CEA-rv)负荷脐血来源树突状细胞(DCs)诱导特异性CIK细胞对结肠癌细胞株Lovo杀伤活性的影响作用。方法取制备好的CEA-rv,用脐血单个核细胞(CBMC)分别制备DCs和CIK细胞;用负荷CEA-rv的DCs和CIK细胞共培养,诱导CEA-rv-DC-CIK细胞。用流式细胞术检测DC免疫表型,MTT法检测CBMC、CIK、Ag-CIK、Ag-DC-CIK和CEA-rv-DC-CIK对Lovo肿瘤细胞株的杀伤活性。结果脐血DCs高表达CD86、CD40,中度表达CD83和CD80。实验组细胞(CEA-rv-DC-CIK)对Lovo细胞的杀伤能力(62%)明显高于对照组(P<0.01)。结论 CEA-rv负荷的DCs能增强CIK细胞对Lovo细胞的杀伤活性,为CEA阳性肿瘤的免疫治疗提供了新的思路。Objective To study the anti-tumor responses of cytokine induced killers to colon cancer cell line Lovo by mature dendritic cells impulsed with CEA-rv derived from cord blood. Methods The dendritic cells and CIK were derived from the same cord blood.Dendritic cells were impulsed with CEA-rv and analyzed by flow cytometry.The anti-tumor effects on Lovo colon cancer cell line by dendritic cells impulsed with CEA-rv mixed with CIK,CIK with DC and CIK were checked with MTT method. Results The morphologically typical dendritic cells derived from cord blood were induced in vitro which can highly expressed costimulatory molecules CD86 CD40 CD80 and CD83.The anti-tumor response to Lovo in experimental group was superior to the others.There was significant difference in rise rate of cytotoxic effect compared with contrast groups. Conclusion Dendritic cells impulsed with CEA-rv could enhance the cytotoxic effect on target cells.This could offer us a new way to CEA positive tumor immunotherapy.
