P16基因和HER2/neu基因在胃癌组织中的表达及其意义

摘　　要：目的研究P16基因和HER2/neu在胃癌中的表达及其临床意义。方法选取未经术前放、化疗的胃癌标本112例及癌旁组织20例,通过免疫组织化学SP法检测两组P16基因和HER2/neu基因的表达情况,分析两者的表达与胃癌临床病理特征的关系。结果胃癌组织P16基因阳性表达率低于癌旁正常黏膜组织,而HER2/neu基因阳性表达率高于癌旁正常黏膜组织;P16的表达与胃癌分化程度和胃癌组织中HER2/neu的表达呈负相关。HER2/neu的表达与胃癌淋巴结转移呈正相关。结论 P16基因的缺失可能在胃癌的发生发展过程中发挥重要作用;HER2/neu的过表达提示预后较差。Objective To investigate the expression and clinical significance of P16 gene and HER2/neu in gastric carcinoma.Methods The expressions of P16 and HER2/neu genes in 112 cases of gastric carcinoma（without radiotherapy or chemotherapy before surgical excision） and 20 cases of carcinoma-adjacent tissues were detected with SP immunohistochemistry.The correlation of the expressions and clinicophathologic characters were analyzed.Results The expression of P16 gene was lower,but the expression of HER2/neu gene was higher,in gastric carcinoma than those in carcinoma-adjacent mucosa tissues.The P16 expression was negatively corretated to the carcinoma differentiation and HER2/neu expression.HER2/neu expression was positively correlated to lymph node metastasis.Conclusion The defect of P16 expression may play a significant role in the occurrence and development of gastric carcinoma.The overexpression of HER2/neu gene may predict a poor prognosis of patients with gastric carcinoma.
