藏药唐古特红景天对急性低氧大鼠脑缺血再灌注损伤的实验研究  被引量:2

摘  要:[目的]探讨藏药唐古特红景天在低氧条件下对大鼠脑缺血再灌注损伤的保护作用和抗疲劳作用。[方法]Wistar大鼠随机分5组,为常氧对照组、低氧组和唐古特红景大、中、小剂量组。将低氧组和唐古特红景大、中、小剂量组放入模拟海拔5500m的低氧低压舱内;常氧对照组和低氧组每天0.9%生理盐水灌胃,红景天每天以成人剂量的20倍、15倍和10倍灌胃,连续给药14d,制脑缺血再灌注损伤大鼠模型,测膈肌最大收缩力、最大强直收缩、强直收缩时间、疲劳时间和疲劳指数以及血清血红蛋白(Hb)、红细胞压积(HCT)、丙二醛(MDA)、超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)、一氧化氮(NO)、内皮素-1(ET-1)的值。[结果]低氧组大鼠的膈肌最大收缩力、最大强直收缩、强直收缩时间、疲劳时间和疲劳指数均较用药组和对照组显著缩短,差异有统计学意义(P﹤0.05),唐古特红景天不同剂量组可显著降低ET-1含量(P﹤0.05),同时MDA、SOD、GSH、GSH-Px和NO与低氧组比较差异也有统计学意义(P﹤0.05)。[结论]藏药唐古特红景天在低氧环境下对脑缺血再灌注损伤具有明显的保护作用。[Objective] To explore Tibetan Rhodiola tanguticum of hypoxic conditions in rat cerebral ischemia-reperfusion injury and anti-fatigue effect.[Methods]Wistar rats were randomly divided into 5 groups for the normoxic control group,hypoxic group and the Red King tanguticum large,medium and low-dose group.The hypoxic group and the Red King tanguticum large,medium and low dose of oxygen were put into the low-voltage analog cabin altitude of 5 500 m;normoxia control group and the hypoxic group took 0.9% saline per day,Rhodiola adult dose 20 times,15 times and 10 times gavage per day,continuous administration for 14 days.We established the system cerebral ischemia rat model,the largest measuring diaphragm contractility,the maximum tonic contraction,tonic contraction time,fatigue time and fatigue index;and serum hemoglobin(Hb),hematocrit(HCT),malondialdehyde(MDA),superoxide dismutase(SOD),glutathione peroxidase(GSH-Px),nitric oxide( NO)and endothelin -1(ET-1)value were detected.[Results]Diaphragm of hypoxic rats the maximum contraction,maximum tetanic contraction,tonic contraction time,fatigue time and fatigue index were significantly reduced than those in treatment group and control group,there was significant difference(P﹤0.05),Tangut Red King days with different doses significantly reduced ET-1 levels(P﹤0.05),while MDA,SOD,GSH,GSH-Px and NO compared with hypoxia group also showed significant differences(P﹤0.05).[Conclusion]Tibetan Rhodiola tanguticum under the hypoxia had protective effects on ischemia reperfusion injury.
