摘 要:为了探讨脱落酸受体ABAR/CHLH是否参与葡萄果实成熟的调控,以玫瑰香葡萄花后8~11周的果实为试材,结合RT-PCR和3’-RACE技术首先克隆了脱落酸受体ABAR/CHLH同源基因。该基因在基因组中以单拷贝形式存在,并编码了1个含有1 381个氨基酸的多肽。生物信息学分析表明,ABAR/CHLH含有3个跨膜区,在进化上高度保守。围绕果实花后8~11周ABAR/CHLH基因转录水平以及ABA含量分析表明,随着果实的退绿,在着色前基因转录水平和ABA含量都迅速增加,此后基因表达量迅速降低,而ABA含量继续缓慢上升至现红后才缓慢降低。因此,玫瑰香葡萄果实的成熟启动不仅依赖于ABA含量快速积累,而且还依赖于其受体基因表达水平迅速增加。To investigate whether ABA receptor ABAR / CHLH is involved in the regulation of grape berry ripening,the ABAR/CHLH gene was first cloned by combination of RT-PCR and 3'-RACE technique with Muscat grape berry during 8 to 11 weeks after anthesis as material.The gene encodes a polypeptide containing 1 381 amino acids,and harbors in genomic DNA with a single copy.Bioinformatics analysis showed that ABAR/CHLH is evolutionarily conserved,and contains three putative transmembrane domains.Analysis of both the transcription level of ABAR/CHLH and ABA content during 8 to 11 weeks after anthesis showed that coupled with de-greening,both levels increased rapidly before red-coloring,thereafter,the former declined rapidly,in contrast,the latter continually and slowly increased up to red-coloring,then began to decline.The results indicated that Muscat grape berry ripening depends not only on the rapid accumulation of ABA contents,but also on the rapid transcription level of its receptor gene.
