摘 要:目的探讨人端粒酶逆转录酶(hTERTmRNA)与突变型p53蛋白在甲状腺癌和甲状腺良性组织中的表达、相关性及临床意义。方法采用原位杂交法检测hTERTmRNA的表达,采用免疫组织化学法检测突变型p53蛋白的表达。结果①hTERTmRNA和突变型p53蛋白在甲状腺癌中高表达(P<0.05)。②hTERTmRNA与p53蛋白阳性率与甲状腺癌的临床病理分型,有无淋巴结转移,肿瘤TNM分期有关(P<0.05),与患者年龄,性别无关。③hTERTmRNA与p53蛋白在甲状腺癌组织中的表达呈显著正相关,具有统计学意义(P<0.05)。结论①hTERTmRNA和突变型p53蛋白的高表达在甲状腺癌的发生发展过程中起到重要作用。②p53蛋白调控hTERTmRNA的表达,p53蛋白在甲状腺癌中表达的增高可激活hTERTm-RNA,从而导致甲状腺癌的发生。③hTERTmRNA和p53蛋白在甲状腺癌中表达的变化可用于评估其生物学行为和判断预后,对临床上甲状腺癌的靶向治疗起到重要的指导作用。Objective To explore the expression,correction and clinical significance of hTERTmRNA and p53 protein in thyroid carcinoma.Methods The expression of hTERTmRNA was detected by in situ hybridization,and the expression of mutant p53 protein was detected by immunohistochemistry.Results ① Overexpression of hTERTmRNA and mutant p53 protein were indicated in thyroid carcinoma tissue(P0.05).② The positive-staining rate of hTERTmRNA and mutant p53 protein were related to the different pathology classification,tumor stages,positive or negative metastasis lymph nodes(P0.05).It was not related to the age and sex of patients(P0.05).③ It demonstrated a significantly positive correlation between the expression of hTERTmRNA and mutant p53 protein in thyroid carcinoma,showing statistical significance(P0.05).Conclusion ① Overexpressed hTERTmRNA and mutant p53 protein may play an important role in the carcinogenesis and progression of thyroid carcinoma.② Mutant p53 protein regulate and control the expression of hTERTmRNA.The expression rising of mutant p53 protein in thyroid carcinoma may activate the expression of hTERTmRNA,resulting in the occurrence of thyroid carcinoma.③ Examination of expression of hTERTmRNA and mutant p53 protein may be helpful in judging the biological behavior and prognosis of thyroid carcinoma.It will perform an important directive function on the gene target therapy of thyroid carcinoma in clinical study.
