摘 要:目的制备培氟沙星与锌配合物,并观察其抗菌活性。方法氯化锌0.136 3 g和培氟沙星0.666 0 g化学反应制备配合物。对配合物进行元素分析和红外光谱分析,并进行体外抗菌活性测定。结果配合物各元素的组成比例为:锌(Zn)13.16%,碳(C)41.05%,氢(H)4.02%,氮(N)8.43%,氯(C l)7.15%,水(H2O)12.50%。由元素分析可推知配合物的分子式为Zn2(C17H20FN3O3)2C l2(H2O)7。培氟沙星在1 713 cm-1处出现的羧基上C=O伸缩振动吸收峰,在配合物中消失,而在1 572.1,1 343.7 cm-1则出现了对应于羧基的不对称和对称伸缩振动吸收峰,培氟沙星在1 634 cm-1出现的萘啶环上羰基(C=O)伸缩振动吸收峰,在配合物中则红移了13个波数至1 621.2 cm-1。培氟沙星对流感嗜血杆菌和粪链球菌的最低抑菌浓度分别为1.25和2.5 mg.mL-1,配合物对流感嗜血杆菌、粪链球菌最低抑菌浓度分别为0.62和1.25 mg.mL-1。结论该配合物制备方法简单,体外抗菌活性结果表明,培氟沙星与锌配合物对流感嗜血杆菌、粪链球菌的抑制作用略高于培氟沙星。Objective To prepare the complex of pefloxacin and zinc,and investigate its antimicrobial activity. Methods The complex was produced by chemical reaction between 0.136 3 g zinc chloride and 0.666 0 g pefloxacin.The elemental analysis and infrared spectroscopy were conducted for the complex detection,and the antibacterial activity was determined in vitro. Results The complex contained 13.16% Zn,41.05% C,4.02% H,8.43% N,7.15% Cl and 12.50% H2O.The molecular formula of the complex was extrapolated to be Zn2(C17H20FN3O3)2Cl2(H2O)7 by the element analysis.The carboxyl C=O stretching vibration absorption peaks appeared in pefloxacin at 1 713 cm-1s,which disappeared in the complexes,and the corresponding asymmetric and symmetric carboxyl stretching vibration absorption peak appeared in pefloxacin at 1 572.1,1 343.7 cm-1.The naphthyridine ring carbonyl(C=O) stretching vibration occurred in pefloxacin at 1 634 cm-1 which in the complexes were red-shifted 13 to 1 621.2 cm-1.The MIC of pefloxacin against Haemophilus influenzae and Streptococcus faecalis MIC were 1.25 and 2.5 mg·mL-1,respectively.While the MIC of the complexes against Haemophilus influenzae and Streptococcus faecalis were 0.62 and 1.25 mg·mL-1,respectively. Conclusion The method for preparation of the complex is simple.It indicates that in vitro antibacterial activity against Haemophilus influenzae and Streptococcus faecalis by the complex of pefloxacin and zinc is slightly higher than that of pefloxacin.
