Ang-2和HIF-1α在膀胱移行细胞癌中的表达及临床意义

摘　　要：目的探讨血管生成素-2(Ang-2)和缺氧诱导因子-1α(HIF-1α)在膀胱移行细胞癌中的表达及其与膀胱移行细胞癌病理分级、临床分期之间的关系,以及两者表达之间的相互关系。方法选取经临床病理诊断的膀胱移行细胞癌标本60例,正常膀胱组织20例。应用免疫组化染色方法检测Ang-2和HIF-1α在膀胱移行细胞癌中的表达情况。结果 Ang-2和HIF-1α在正常膀胱组织中均无表达;在膀胱移行细胞癌标本中均呈过表达。膀胱移行细胞癌中Ang-2和HIF-1α的阳性表达率分别为47.7%、41.7%。膀胱移行细胞癌中,Ang-2和HIF-1α阳性表达率随其病理分级和临床分期增加而升高。经统计检验,Ang-2与HIF-1α的等级相关系数rs为0.498,P<0.01,差异有统计学意义。结论膀胱组织在癌变过程中获得了Ang-2、HIF-1α受体。Ang-2、HIF-1α的阳性表达率均随膀胱移行细胞癌的病理分级、临床分期增加而升高,表明Ang-2、HIF-1α阳性率越高,膀胱移行细胞癌的恶性度越高,浸润越深。膀胱移行细胞癌中Ang-2和HIF-1α的表达具有两两正相关关系;两者相互作用,通过影响肿瘤血管形成,共同参与膀胱移行细胞癌的发生、发展。Objective To investigate the relationships among the expression of angiopoietin-2（Ang-2）,hypoxia-inducible factor 1α（HIF-1α） and the clinicopathological variables in patients with transitional cell carcinoma（TCC） of the urinary bladder,the correlation of Ang-2 and HIF-1α in transitional cell carcinoma of urinary bladder.Methods The expression of Ang-2 and HIF-1α were assessed by immunohistochemistry in tissue sections of 60 patients with transitional cell carcinoma of the urinary bladder and controloed with that of 20 normal bladder samples.Results There was no marked expression of Ang-2 and HIF-1α in normal bladder samples.The positive expression of Ang-2 and HIF-1α was detected in TCC samples.The positive rate of Ang-2 and HIF-1α was 47.7%（28/60） and 41.7%（25/60）,respectively.In TCC,positive Ang-2 and HIF-1α expression was significantly correlated with histological grade,clinical stage.There were relatively correlations in the overexpression of Ang-2 and HIF-1α in TCC each other（rs=0.498,P0.01）.Conclusion These results suggest that bladder tissue had got Ang-2 receptor and HIF-1α receptor in cancerization.The overexpression of Ang-2 and HIF-1α are associated with tumour grades and stages.
