摘 要:目的研究出生前后不同浓度慢性铝暴露对年轻大鼠学习记忆行为学及海马细胞内CaMKⅡ表达影响,以探讨铝损害学习与记忆的突触机制。方法 45只大鼠随机分为对照组、0.2%-Al和0.4%-Al组大鼠,从孕期始分别自由饮用蒸馏水、Al3+浓度为15mmol·L-1和30mmol·L-1的A1C13水溶液;采用跳台法测定海马长时程增强,采用Western-blot法检测海马细胞内CaMKⅡ的表达。结果大鼠记忆保持测试结果显示,与对照组相比,各铝暴露组跳下平台的潜伏期明显缩短(P<0.01),5min内错误次数显著增加(P<0.01),且两铝暴露组间差异明显(P<0.05);与对照组相比,两铝暴露组海马CaMKⅡ的表达明显降低(P<0.01)。结论出生前后慢性铝暴露引起大鼠学习记忆能力的降低可能与海马CaMKⅡ的表达下调相关。Objective To study the change of CaMKⅡ in hippocampus of young rats after chronic aluminum exposure of different concentrations from prenatal to postnatal to investigate the damaged mechanism of learning and memory capacity caused by aluminum.Methods 45 rats were randomly divided into:normal control group,0.2%-Al group(15mmol·L-1 Al in drinking water),0.4%-Al group(30mmol·L-1 Al in drinking water) Learning and memory ability was tested by step down test,the expression of CaMKⅡin hippocampus was measured by Western-blot.Results Compared with the control group,latency period was significantly extend in aluminum exposure group by step down test,the error number was increased in 5minutes.The learning and memory ability was damaged in Al3+ exposed groups(P〈0.01)with more serious in 0.4%-Al group than in 0.2%-Al group(P〈0.05).The expression of CaMKⅡ was decreased in aluminum exposure group than in normal control group(P〈0.01).Conclusion The damaged learning and memory ability by aluminum exposure from prenatal to postnatal might be related to the expression of CaMKⅡ.
