摘  要:目的研究缓激肽(bradykinin,BK)对脑胶质瘤大鼠肿瘤微血管内皮细胞中转录因子核因子-κ B(nuclearfactor-kappaB,NF-κ B)的活性和表达的影响。方法雌性Wistar大鼠96只,随机分为6组,即对照组、BK 5min组、BK 10min组、BK 15min组、BK 30min组和BK 60min组。每组16只。建立大鼠C6胶质瘤模型,颈动脉给药,应用Trans AMTMNF-κ B p65试剂盒检测NF-κ B(p65)的活性;应用免疫组织化学法检测大鼠脑肿瘤微血管上NF-κ B(p65)的分布和表达水平变化;应用western blot法检测肿瘤微血管内皮细胞中NF-κ B(p65)蛋白表达水平的变化。结果与对照组相比,BK显著增强了NF-κ B的活性;在脑肿瘤组织,NF-κ B(p65)主要表达在肿瘤微血管和肿瘤细胞上,BK能够增加NF-κ B(p65)在肿瘤微血管上的表达,并且上调肿瘤微血管内皮细胞细胞核中NF-κ B(p65)的蛋白表达,在BK作用15min时效果最显著(P<0.01)。结论缓激肽能够增强脑胶质瘤大鼠肿瘤微血管中NF-κ B的活性和表达水平,NF-κ B可能是BK开放血肿瘤屏障机制之一。Objective To investigate the effect of bradykinin (BK) on the activity and expression of nuclear factor-kappa B (NF-кB) in microvascular endothelial cells of brain glioma rat.Methods 96 female rats were divided averagely into 6 groups:control,BK 5min group,BK 10min group,BK 15min group,BK 30min and BK 60min groupd.The C6 brain glioma rat model was established and BK was pumped into rats,brain via the carotid artery,trans AMTM NF-κB p65 Kit was used to measure the activity of NF-kB.The distribution and expression of NF-кB (p65) in tumor microvessels were detected by SP immunohistochemistry and Western blot respectively.Results In contrast to control group,BK obviously enhanced the activity of NF-кB (p65).In glioma rats,model,NF-кB (p65) mainly expressed in tumor microvessels and tumor cells,and the expression level of NF-кB (p65) in tumor microvessels was increased after BK infusion compared with glioma rats,model.Moreover,BK could upregulate the expression of NF-кB (p65) protein in the nucleus of tumor microvascular endothelial cells and reach peak at BK infusing for 15min (P〈0.01).Conclusions BK could increase the activity and expression of NF-кB (p65) in microvessels of brain glioma rats,NF-кB might be involved in the mechanism of BK opeining the brain tumor barrier.
