摘 要:目的探讨肾小球基质金属蛋白酶-2(MMP-2)的表达与微小病变型肾病综合征的关系。方法以雄性SD大鼠为研究对象,通过尾静脉注射阿霉素的方法建立肾病综合征模型,对照组注射生理盐水。通过透视电镜观察肾小球的病理变化;运用免疫组织化学方法(SABC法)检测肾小球中MMP-2的表达;运用LEICAQWIN图像信号采集与分析系统对免疫组织化学图像进行分析处理。结果造模后阿霉素肾病模型组大鼠肾小球基底膜薄厚不均,足突广泛融合,间隙消失,排列紊乱,符合微小病变型肾病综合征的病理改变;模型组大鼠24 h尿蛋白定量明显高于对照组(P<0.01),模型组大鼠肾小球MMP-2表达高于对照组(P<0.01)。结论基质金属蛋白酶-2参与微小病变型肾病综合征肾小球足突细胞的损伤。Objective To investigate the relationship between the expression of matrix metalloproteinases-2 (MMP-2) in glomerulus and minimal change nephritic syndrome. Methods Male SD rats were subjected to the experiment. The nephropathy model was established by giving a single vena caudalis injection of adriamycin, while the normal group received injection of the same-dose sodium chloride. We observed the pathological changes of glomerulus through perspective electron microscope; immunohistochemical method (SABC) was used to detect MMP-2 expression in glomerulus. At last, we used LEICAQWIN image signal acquisition and analysis system to process these data. Results With electron microscope, we observed that the thickness of glomerular basement membrane in model group was uneven, the foot processes generally effaced and foot gaps disappeared, which coincided with the pathological features of minimal change nephritic syndrome. In model group, the 24h urine protein was obviously higher than that in normal group (P〈0.01) and MMP-2 was more highly expressed than in normal group (P〈0.01). Conclusion The expression of MMP-2 is involved in glomerular foot cell injury in minimal change nephritic syndrome.
