多西他赛节律化疗对胃癌细胞及内皮细胞生物学效应影响的观察

摘　　要：目的:探讨多西他赛节律化疗对胃癌细胞及内皮细胞生物学效应的影响。方法:采用低剂量的多西他赛持续作用于人脐静脉内皮细胞株(HUVEC)和人BGC-823胃癌细胞株144h,观察细胞增殖、凋亡和血小板反应素1(TSP-1)的表达情况。结果:多西他赛对人HUVEC较BGC-823胃癌细胞的抑制作用更强,IC50值分别为(0.06±0.005)和(1.1±0.024)nmol/L。较低IC50浓度的多西他赛作用于HUVEC引起细胞凋亡的百分比近似于较高IC50浓度的多西他赛作用于BGC-823细胞的百分比。多西他赛明显地增加了人HUVEC的TSP-1表达,为对照组的(1.78±0.18)倍,而且明显地增加了TSP-1蛋白的分泌,与对照组的100%相比,多西他赛组为(806±73)%(P<0.05),而肿瘤细胞的TSP-1表达和分泌没有增加。结论:多西他赛节律化疗优先抑制了内皮细胞的增殖和诱导内皮细胞的凋亡,其机制可能是由于多西他赛节律化疗增加了内皮细胞TSP-1的表达和分泌。OBJECTIVE： To investigate the effects of met- ronomic docetaxel chemotherapy on biological behaviors of gastric cancer cells and endothelial cells. METHODS： In vitro cell proliferation, apoptosis and thrombospondin-1/ TSP-1 expression analyses were performed on human umbilical vein endothelial cells （HUVEC） and gastric cancer cells （BGC-823）, exposed for 144 h to metronomic concentrations of docetaxel. RESULTS： HUVEC was more sensitive to docetaxel （IC50 （0.06± 0. 005） nmol/L] than BGC-823 cells （IC50 （ 1.1± 0. 024） nmol/L]. The percentage of apoptotic HUVEC at lower IC50 levels was similar to that obtained, when treating BGC-823 cells at higher IC50 levels. Doeetaxel significantly increased TSP-1 expression [（1. 78±0.18） times of that of controls] and secretion [（806±73）% vs 100% of controls] in HUVEC. In contrast, TSP-1 expression and secretion were not increased in BGC-823 cancer cells. CONCLUSIONS： Metronomic docetaxel treatment preferentially inhibits endothelial cell proliferation and induces endothelial cell apoptosis. These findings may be explained by the changes of TSP-1 expression and secretion.
