复方苦参注射液对人胃癌BGC-823细胞株增殖抑制作用研究

摘　　要：目的探讨复方苦参注射液对人胃癌BGC-823细胞株增殖的抑制作用。方法采用四甲基偶氮唑蓝(MTT)比色法检测复方苦参注射液对人胃癌BGC-823细胞48 h增殖抑制作用,并求出其48 h半数抑制浓度(IC50);用流式细胞仪分析技术及琼脂糖凝胶电泳检测复方苦参注射液对人胃癌BGC-823细胞凋亡的影响。结果复方苦参注射液能明显抑制人胃癌BGC-823细胞增殖并呈浓度依赖性,IC50为9/250原液稀释倍数;人胃癌BGC-823细胞经复方苦参注射液(终浓度为1/25.0原液稀释倍数)作用24 h、48 h及72 h后,流式细胞仪分析结果显示,其早期凋亡细胞率分别为(9.58±0.88)%、(20.41±0.77)%及(60.30±3.19)%,明显高于空白对照组的(4.17±0.11)%,差异均有统计学意义(P<0.05),并发现明显的细胞凋亡峰(Sub-G1);人胃癌BGC-823细胞经复方苦参注射液(终浓度为1/25.0原液稀释倍数)作用48 h和72 h后,可见明显的梯状条带形成。结论复方苦参注射液可抑制人胃癌BGC-823细胞增殖,并诱导其凋亡。Objective To explore the inhibition effects of Compound Radix Sophorac Flavescentis Injection（CRSFI） on proliferation of human gastric cancer BGC-823 cells.Methods MTT assay was used to determine the effect of 48-hour CRSFI incubation on the proliferation of human gastric cancer BGC-823 cells,with the concentration required for 50% growth inhibition（IC50） calculated.Apoptosis status of the cells were investigated through flow cytometry and agarose gel electrophoresis.Results MTT analysis indicated that proliferation of human gastric cancer BGC-823 cells were inhibited markedly by CRSFI in a dose-dependent manner.IC50 for 48-hour incubation was 9/250 of the original fluid.Flow cytometry showed that CRSFI（dilution：1/25.0） incubation of 24 h[（9.58±0.88）%],48 h[（20.41±0.77）%],and 72 h[（60.30±3.19）%] led to enhanced apoptotic rates compared to the control group[（4.17±0.11）%] in human gastric cancer BGC-823 cells（P0.05）,with Sub-G1 peaks noticed.DNA ladder formation was also noticed in BGC-823 cells incubated with CRSFI（dilution：1/25.0）for both 48 and 72 hours in a agarose gel electrophoresis assay.Conclusion CRSFI exhibits a effect of proliferation inhibition and apoptosis induction in human gastric cancer BGC-823 cells.
