摘 要:目的体外、原位实验观察HRP/IAA酶前药系统对鼻咽癌C666-1细胞的杀伤效应,以探讨其对鼻咽癌的治疗作用。方法细胞转染获得稳定表达HRP蛋白的C666-1细胞。MTT法检测不同浓度的IAA对HRP表达阳性的C666-1细胞的杀伤效应。原位基因治疗:取5×106对数生长期的C666-1细胞接种裸鼠右腋下,建立鼻咽癌裸鼠移植瘤模型,接种细胞10d后将裸鼠随机分成3组,观察HRP/IAA酶前药系统对移植瘤生长的抑制作用。结果体外实验中,同一浓度IAA干预后C666-1/HRP(+)组细胞的相对存活率显著低于C666-1/HRP(-)组(P<0.05),且随着IAA浓度的升高C666-1/HRP(+)组细胞的相对存活率逐渐降低,分别为(15.58±1.12)%、(10.11土1.06)%、(5.21±1.04)%;原位基因治疗实验结果表明IAA组鼻咽癌移植瘤生长受到抑制,体积明显小于空病毒组和生理盐水组(P<0.05),且肿瘤细胞凋亡显著增多(P<0.05)。结论 HRP/IAA酶前药系统对鼻咽癌C666-1细胞具有杀伤作用,具有抗鼻咽癌的作用。Objective Observe the damage effect of HRP /IAA enzyme /pro-drug system on nasopharyngeal carcinoma(NPC) C666-1 cells in the in vitro and in situ experiments to investigate the therapeutic effect on NPC.Methods Cells were transfected to obtain the stable HRP gene expression in C666-1 cells.The damage effect of different concentrations IAA on HRP positive C666-1 cells was detected by MTT assay.Furthermore,a NPC model was employed by inoculating C666-1 cells in the right flank of nude mice for in situ gene therapy,and after 10 days of inoculation,those rats were randomized to 3 groups,then the suppression of NPC growth in model was observed after giving different treatments.Results In vitro study showed that the relative survival rates of HRP-expressing C666-1 cells treated with IAA were lower than no-HRP-expression C666-1 cells(P 0.05).And the relative survival rate of C666-1 /HRP(+) group was gradually decreased along with the IAA concentration increases:(15.58 ±1.12)%,(10.11 ±1.06)% and(5.21 ±1.04)%.In situ,potent growth inhibition on the explanted NPC tumors was observed in mice treated with IAA.The tumors volumes in group interfered by HRP was smaller than the null group and the NS group(P 0.05),and the number of the apoptotic cells was more(P 0.05).Conclusion C666-1 cells or nasopharyngeal carcinoma could be killed by HRP /IAA enzyme /pro-drug system,showing a therapeutic effect on NPC.
