摘 要:目的探讨不同刺激因素对垂体腺瘤中血管内皮生长因子(VEGF)分泌的影响。方法原代培养垂体腺瘤细胞,给予地塞米松(10-6 mol/L)、溴隐亭(10-6 mol/L)、白细胞介素-6(IL-6,10 ng/ml)、垂体腺苷酸环化酶激活多肽-38(PACAP-38,0.1 mol/L)、奥曲肽(10-7 mol/L)等外源性刺激,应用酶联免疫吸附试验(ELISA)检测垂体腺瘤细胞中VEGF的分泌量,并比较刺激前后VEGF分泌的变化。结果人垂体腺瘤可检测到VEGF分泌,其中90%垂体腺瘤中VEGF分泌受地塞米松显著抑制。生长激素腺瘤给予PACAP-38刺激后,VEGF分泌水平升高(P<0.05)。结论地塞米松抑制垂体腺瘤细胞VEGF的分泌,说明糖皮质激素具有抗血管形成特性,可用于垂体腺瘤治疗。Objective To investigate the influence of different stimuli on vascular endothelial growth factor(VEGF) secretion in pituitary adenomas.Methods Primary adenomas cells were cultured and stimulated by dexamethasone(10-6mol/L),bromocriptine(10-6mol/L),interleukin-6(IL-6,10ng/ml),pituitary adenylate cyclase-activating polypeptide-38(PACAP-38,0.1mol/L) and octreotide(10-7mol/L).Enzyme-linked immunoassay(ELISA) was employed to detect the secretion of VEGF in the pituitary adenoma cells,and the amounts secreted before and after stimulation were compared.Results The secretion of VEGF could be detected in pituitary adenomas and inhibited by dexamethasone in 90% pituitary adenomas.The secretion was increased significantly in growth hormone adenomas after the stimulation with PACAP-38(P0.05).Conclusions Dexamethasone can inhibit the secretion of VEGF in pituitary adenoma cells,which indicates that glucocorticoids possess anti-angiogenesis effect and can be used to treat pituitary adenomas.
