摘  要:目的:观察凋亡相关基因Bcl-2、核因子NF-κB在肝细胞性肝癌(HCC)组织中的表达,并探讨二者与AFP之间的关系。方法:采用免疫组织化学方法检测40例HCC组织(其中AFP阳性20例,AFP阴性20例)和10例良性肿瘤组织中Bcl-2、NF-κB的表达。结果:40例HCC患者Bcl-2阳性表达率为45.0%,NF-κB阳性表达率为57.5%;其中AFP阳性细胞Bcl-2表达率为55.0%,AFP阴性细胞为35.0%;AFP阳性细胞NF-κB表达率为75.0%,AFP阴性细胞为40.0%。HCC细胞NF-kB和Bcl-2的表达呈正相关(r=0.778,P<0.01)。结论:Bcl-2、NF-κB的异常表达同HCC的抗凋亡和耐药性有一定关系,AFP阳性HCC细胞的Bcl-2、NF-κB阳性表达率更高。Bcl-2和NF-κB可以考虑作为预后的判断指标之一。Objective:To observe the apoptosis-related gene Bcl-2,nuclear factor NF-κB expression in hepatocellular carcinoma cells and to explore the relationship with AFP and its clinical significance.Methods:Immunohistochemical technique was used to detect the expression of Bcl-2,NF-κB in 40 cases of hepatocellular carcinoma(including 20 cases of AFP-positive,20 cases of AFP-negative) and 10 cases of benign tumor.The relationship between them and AFP was ananlyzed with Spearman correlation test with spss13.0.Results:The expression rate of Bcl-2 was 45%,NF-κB expression rate was 57.5% in 40 cases;in which expression of Bcl-2 in AFP-positive cells was 55%,AFP-negative cells was 35%;NF-κB expression in AFP-positive cells was 75%,in AFP negative cells was 40%.The expression of NF-kB protein and Bcl-2 protein was significantly positively correlated(r=0.778,P〈0.01) in hepatoma carcinoma cells.Conclusion:The abnormal expression of Bcl-2 and NF-κB have a certain relationship with tumor resistance to apoptosis and drug resistance,The expression rate of Bcl-2 and NF-κB is higher in AFP-positive cancer cells.Bcl-2 and NF-κB can be considered as a prognostic indicator.
