蒿鳖养阴软坚方给药大鼠体内乙醛含量的测定

摘　　要：目的研究建立蒿鳖养阴软坚方给药大鼠血浆中乙醛浓度的HPLC测定方法。方法色谱柱Hypersil C18(250 mm×4.6 mm,5μm);流动相为乙腈-水,梯度洗脱(起始浓度5%,10 min15%,20 min40%),流速1.0 mL/min;柱温:30℃,检测波长:210 nm,进样量:10μL。结果乙醛在线性范围0.0025～0.005μL/mL之间线性关系良好(r=0.999 6)。测定的方法回收率是80.4%(RSD=1.78%)。结论该测定方法简单可靠。Objective To establish an HPLC method for determination of acetaldehyde level in rat plasma. .＇Methods HPLC analysis was performed on the Hypersil C18 column （250 mm×4.6 mm,5 μm） at temperature 30 ℃,The mobile phase consisted of acetonitrile and water,using gradient elution （5% at the begining, 15% at 10 rain, 40% at 20 min ） with the flow rate 1,0 mL·min^-1,The deteetion wavelength was 210 nm. Results The statldard curve was linear in the range of 0.002 5-0.005μL/mL （r =0.999 3）.The method recovery was 80.4%, with RSD=1.78% .Conclusion The method is convenient and sensitive.
