摘 要:目的:观察黄芪注射液对阿霉素中毒性心肌损伤模型(ADR)小鼠心肌线粒体膜电位及还原型谷胱甘肽(GSH)的影响。方法:取昆明小鼠60只,随机分为黄芪注射液治疗组(大、中、小3个剂量组)、阿霉素模型组和正常对照组,建立ADR,实验结束24h后处死小鼠,测定心脏系数和心肌组织中GSH的含量;应用流式细胞仪分析技术检测小鼠心肌细胞线粒体膜电位。结果:黄芪注射液各剂量组心脏系数与阿霉素模型组比较均显著降低(P<0.01);黄芪注射液各剂量组分别与阿霉素模型组比较GSH含量均增高(P<0.01);黄芪注射液各剂量组间心脏系数和GSH比较差异无统计学意义(P>0.05)。线粒体膜电位黄芪注射液各剂量组较阿霉素模型组明显增高(P<0.05或P<0.01),黄芪注射液各剂量组与正常对照组比较差异也有统计学意义(P<0.05)。结论:黄芪注射液可通过减轻小鼠心脏肿大,提高心肌内GSH含量,稳定线粒体膜电位,抑制心肌细胞发生异常凋亡来实现其保护心肌细胞的作用。Objective:To observe the effect of Huangqi injection on the mitochondrial membrane potential(MMP) and reduced glutathione hormone(GSH) in mice.Methods:Sixty Kunming mice were randomly divided into five groups,normal control group,adriamycin(ADR) group and Huangqi injection group(high-dose,middle-dose and low-dose).Mouse models of myocardial injury were established by toxicity of doxorubicin.The values of the coefficient of the heart and GSH were detected,and MMP was tested by flow cytometry.Results:Compared with ADR group,there was a lower value of the coefficient of the heart in Huangqi injection group(P 〈 0.01).The level of GSH was higher in Huangqi injection group than that of ADR group(P 〈 0.01).There were no significant differences between the high-dose,middle-dose and low-dose of Huangqi injection groups(P 〉 0.05).The value of MMP was significantly higher in Huangqi injection group than that of ADR group(P 〈 0.01),and there was a significant difference between Huangqi injection group and normal control group(P 〈 0.05).Conclusion:Huangqi injection protects cardiac muscle cells by relieving heart tumefy,increasing the level of GSH,stability MMP and restraining the apoptosis of cardiac muscle cells.
