蒿鳖养阴软坚方对ConA诱导小鼠肝纤维化的预防作用

摘　　要：目的:探讨蒿鳖养阴软坚方对伴刀豆球蛋白A(ConA)诱导小鼠肝纤维化的预防作用。方法:94只Balb/c小鼠根据体质量采用随机区组法分为7组,正常组8只,模型组16只,蒿鳖养阴软坚方高、中、低剂量组,复方鳖甲软肝片组和秋水仙碱组各14只。采用尾静脉注射ConA制备小鼠肝纤维化模型,以蒿鳖养阴软坚方作为预防用药。采用HE染色及胶原染色法观察肝组织形态;盐酸水解法测定肝组织胶原蛋白的含量;放射免疫法测定血清透明质酸(HA)和Ⅲ型前胶原(PCⅢ)水平。结果:模型组肝纤维化程度分期及计分均高于正常组(P<0.01),中、低剂量组肝纤维化程度分期均低于模型组(P<0.05)。模型组肝组织胶原蛋白含量高于正常组(P<0.01),高、中、低剂量组肝组织胶原蛋白含量皆明显低于模型组(P<0.05或P<0.01)。结论:蒿鳖养阴软坚方对ConA诱导小鼠肝纤维化具有预防作用。Objective：To investigate the prophylactic effect of HaoBieYang YinRuanJian（HBYYRJ） herbal decoction on liver fibrosis induced by concanavalin A（ConA）in mice.Methods：Ninety four Balb/c mice were randomly divided into 7 groups,including normal group（n=8）,model group（n=16）,HBYYRJ herbal decoction treatment groups（low dosage,middle dosage and high dosage,n=14 for each group）,composed formula treatment group（n=14） and colchicine treatment group（n=14）.Mouse models of hepatic fibrosis were established by injection of ConA.HBYYRJ was used for prevention of hepatic fibrosis.Histological observation of the liver tissue was detected by H＆E stain and collagen stain.The collagen？content of？liver tissue was detected by hydrochloric acid？hydrolysis？method.The levels of serum hyaluronic caid（HA） and procollagen type Ⅲ（PC Ⅲ） were examined by radioimmunoassay.Results：The degree of liver fibrosis staging was significantly higher in model group than that in the normal contrast group（P 〈 0.01）.In the three groups of HBYYRJ,the degree of hepatic fibrosis was significantly lower in low and middle dosage groups than that in the model group（P 〈 0.05）.The content of collagen was significantly higher in the model group than that of normal control group（P 〈 0.01）.The values of collagen content were significantly lower in the HBYYRJ（high medium and low dosage） groups than that in the model group（P 〈 0.05）.Conclusion：HBYYRJ herbal decoction showed preventive effect on the liver fibrosis induced by ConA in mice.
