舌鳞状细胞癌对卡铂耐药过程中切除修复交叉互补基因的表达

摘　　要：目的探讨Tca8113对卡铂（carboplatin）体外耐药过程中，切除修复交叉互补基因1（excision repair cross-complementation 1，ERCC-1）的表达与癌细胞耐药的关系。方法采用逐步递增法将不同质量浓度卡铂体外作用于Tca8113，设置1个对照组并筛选出6个耐药组（1～6组）；采用甲基噻唑基四唑（MTT）法检测每组细胞的半数抑制浓度（half inhibitory concentraction，IC50），反转录聚合酶链反应法（RT-PCR）和蛋白质印迹法检测每组细胞ERCC-1在mRNA及蛋白水平的表达。结果随着耐药性的逐渐增加，各组细胞ERCC-1在mRNA水平的表达逐渐增加（对照组0．84±0．06，1组0．96±0．06，2组1．07±0．11，3组1．13±0．09，4组1．19±0．08，5组1．29±0．07，6组1．47±0．08），相邻或间隔数组间比较，差异有统计学意义（P〈0．05）；各组细胞ERCC-1在蛋白水平的表达逐渐增加（对照组0．69±0．05，1组0．86±0．04，2组1．01±0．04，3组1．24±0．09，4组1．44±0．14，5组1．56±0．19，6组1．88±0．22），相邻或间隔数组间比较，差异有统计学意义（P〈0．05）；各组细胞IC50值与RT—PCR条带灰度值呈正相关（r=0．91，P〈0．01）；与蛋白质印迹条带灰度值呈正相关（r=0．88，P〈0．01）。结论ERCC-1的高表达可能为Tca8113对卡铂耐药的一个标志；为反转耐药提供了一个可能的作用靶点。Objective To investigate the correlations between the resistant degree of Tca8113 agaist carboplatin and the expression of excision repair cross-complementation 1 （ ERCC-1 ）. Methods Tca8113 cells were exposed in the carboplatin solutions of gradually increasing concentration. Cancer cells were divided into six experimental groups and a control group. In the course of culture, the half inhibitory concentration （ICs0）of each groups were detected by methyl thiazolyl tetrazolium（MTT）. The expressions of ERCC-1 at mRNA and protein level were detected by reverse transcription polymerase chain reaction （RT-PCR） and Western blotting. Results With the increase of drug resistance , the expression of ERCC-1 increased gradually at mRNA level（control group：0. 84±0. 06, group 1：0. 96 ±0. 06,group 2.1.07 ±0. 11 ,group 3 ： 1.13 ± 0.09, group 4：1.19 ± 0.08, group 5：1.29 ± 0.07, group 6. 1.47 ± 0.08）,values between neighboring or interval groups have significant differences （P 〈 0. 05 ） ;the expression of ERCC-1 increased gradually at protein level （ control group ： 0. 69 ± 0. 05, group 1 ： 0. 86 ± 0. 04, group 2 ： 1.01 ±0. 04, group 3 ： 1.24 ± 0.09, group 4：1.44 ± 0.14, group 5：1.56 ± 0.19, group 6：1.88 ± 0.22）,values between neighboring or interval groups have significant differences （ P 〈 0. 05 ）. The gradually increasing RT-PCR gray values were related to the gradually increasing ICso in all groups （r = 0. 91, P 〈 0. 01 ） , and the gradually increasing western blotting gray values were also related to the gradually increasing ICs0 in all groups （r = 0. 88,P 〈 0. 01）. Conclusions The high expression of ERCC-1 may become an indicator of Tca8113 resisting carboplatin. This might provide a potential target to reverse tongue squamous cell cancer（ Tca8113 ） that has resistance to carboplatin.
