肝细胞癌c-fos的表达及其与乙型肝炎病毒的关系

摘　　要：目的　研究肝癌组织中ｃ－ｆｏｓ蛋白和ｃ－ｆｏｓｍ ＲＮＡ 的表达及其与乙型肝炎病毒（ＨＢＶ）的关系。　方法　采用免疫组化及原位杂交技术检测５９ 例肝细胞癌组织及５６ 例癌旁组织中ＨＢｓＡｇ、ＨＢＶ ＤＮＡ、ｃ－ｆｏｓ蛋白及ｃ－ｆｏｓｍ ＲＮＡ的表达。　结果　癌旁肝组织ＨＢｓＡｇ 阳性率为８７．５％ （４９／５６）。肝癌组织ＨＢＶ ＤＮＡ阳性率为５９．３％ （３５／５９）。肝细胞癌组织、癌旁肝组织ｃ－ｆｏｓ蛋白的阳性率分别为３９．０％ 和５３．６％ ，ｃ－ｆｏｓｍ ＲＮＡ 的阳性率分别为３３．９％ 和５０．０％ 。癌旁肝细胞不典型增生组ｃ－ｆｏｓ蛋白、ｃ－ｆｏｓｍ ＲＮＡ 的阳性率均显著高于非不典型增生组（Ｐ＜ ０．０５），非癌正常对照组均为阴性。癌组织ＨＢＶ ＤＮＡ 阳性组病例ｃ－ｆｏｓ蛋白、ｃ－ｆｏｓｍ ＲＮＡ 的阳性率显著高于癌组织ＨＢＶＤＮＡ 阴性组（Ｐ＜ ０．０５）。　结论　肝细胞癌组织中存在ｃ－ｆｏｓ癌基因的异常表达，并与ＨＢＶ ＤＮＡ有密切关系。To study the expression of c fos in protein and mRNA levels in hepatocellular carcinoma(HCC) and its association with HBV \ Methods\ Using immuno histochemistry and in situ hybridization to detect HBsAg,HBV DNA,c fos protein and c fos mRNA in carcinomas tissue of 59 cases HCC and 56 paratumorous liver tissue \ Results\ Positive rates of HBsAg in paratumorous liver tissue and HBV DNA in tumor tissue were 87 5%(49/56) and 59 3%(35/59) respectively \ Positive rates of c fos protein in tumor tissue and surrounding liver tissue were 39 0% and 53 6% respectively \ Positive rates of c fos mRNA in tumor tissue and surrounding liver tissue were 33 9% and 50 0% respectively \ The expression of c fos protein and c fos mRNA was increased significantly in dysplastic liver cells (DLC) of paratumorous liver tissue compared with those of non dysplastic liver cells(NDLC) (P<0 05) \ c fos protein and c fos mRNA were detected in HCC more frequently in patients whose tumor tissues had detectable HBV DNA than in those tumor tissues without detectable HBV DNA(P<0 05) \ Conclusion\ There was overexpression of c fos oncogene in HCC,which was closely associated with HBV DNA \;
