摘  要:目的:探讨IL-15基因修饰的自然杀伤细胞系(简称NK-ustc细胞系)对荷人卵巢癌腹水瘤裸鼠模型的治疗效果。方法:将不同剂量的人卵巢癌细胞注入裸鼠腹腔,观察裸鼠的成瘤时间、成瘤率,确定建立腹水瘤模型的最佳剂量;腹水瘤模型建立后,随机分为治疗组(荷瘤鼠腹腔注射照射后的NK-ustc细胞)和对照组(荷瘤鼠腹腔注射无血清α-MEM培养基),同时设空白对照组(裸鼠腹腔注射照射后的NK-ustc细胞),分别观察各组裸鼠腹围改变及生存时间。结果:腹腔注射2.0×106cells/0.2 m l人卵巢癌HO-8910细胞,裸鼠成瘤时间为(28±5)天,成瘤率为100%;使用照射后的NK-ustc细胞治疗荷腹水瘤裸鼠,治疗组裸鼠的腹围低于对照组,但差异无统计学意义(P>0.05);观察到90天,治疗组中位生存期为54天,对照组裸鼠中位生存期为27天,两组比较,差异有统计学意义(P<0.01),空白对照组全部存活,生长良好。结论:使用NK-ustc细胞治疗卵巢癌腹水瘤裸鼠模型是安全、有效的,为卵巢癌的细胞治疗提供了理论依据。Objective:To discuss the anti-tumor activity of gene-modified human natural killer cells(NK-ustc cells) against the tumor-bearing nude mice.Methods:Female BALB/c nude mice were intraperitoneally injected different numbers of human ovarian cells,the model of nude mice bearing ovrian ascitic tumor were determined by the time and rate of tumor formation.The tumor-bearing nude mice were randomly divided into the experimental group(intraperitoneal injection of irradiated NK-ustc cells) and positive control group(intraperitoneal injection of serum-free α-MEM medium).At the same time,as negative control group,only irradiated NK-ustc cells were injected the nude mice.The injected mice were monitored twice weekly for the abdomen circumference and the survival.Results:HO-8910 cells(2×106) were injected into the nude mice,and the mice developed palpable tumor on(28±5) days after tumor cells implantation with the 100% tumorigenesis incident.The tumor-bearing nude mice were treated with irradiated NK-ustc cells,the abdomen circumference of the experimental group were less than positive control group.The difference had no statistical significance(P0.05).Until the 90 day under observation,the median survival of the experimental group was 54 days,though the positive control group was 27 days.The difference of the two groups had statistical significance(P0.01).The negative control group were all survival,growing well.Conclusion:NK-ustc cells show potent anti-tumor activity in nude mice model and provide a theoretical basis for human ovarian cancer cellular therapy.
