膀胱癌组织中Plk1的表达和临床意义

摘　　要：目的:探讨Plk1蛋白在正常膀胱组织和膀胱癌中的表达,及其与临床分期和病理分级的关系。方法:应用western blot和链霉菌抗生物素蛋白-过氧化物酶连接(SP)法检测21例正常膀胱组织、90例膀胱癌组织中Plk1的表达情况及其与膀胱癌各临床病理参数的关系。结果:western blot显示膀胱癌和癌旁正常组织中均有Plk1的表达,但在膀胱癌中的表达量明显高于癌旁正常组织的表达量且具有统计学意义。免疫组化显示膀胱癌组织中Plk1蛋白表达总阳性率为64.44%(58/90),显著高于正常膀胱组织的38.10%(8/21),而且与肿瘤病理分级及肿瘤临床分期具有显著的相关性。结论:Plk1蛋白的阳性表达与膀胱癌的发生、发展密切相关,可以为膀胱癌的良、恶性分析及判断肿瘤的生物学行为提供理论依据。Objective： To detect the expression of Plk1 and to discuss their clinical significance in normal bladder tissue and the bladder cancer.Methods： The expression of Plk1 was detected in 90 bladder cancer tissues and 21 normal bladder mucosa tissues by immunohistochemical streptavidin-perodidase（S-P） method and Western blot.Results： Western blot showed that there was the expression of Plk1 in normal bladder tissues and bladder tumors,but the expression in bladder tumors was significantly higher than that in normal bladder tissues.Immunohistochemistry showed that the expression rate of Plk1 in the bladder cancer tissues 64.44 %（58/ 90） was significantly higher than that in normal bladder tissues 38.10 %（8/21）.Moreover the expression of Plk1 was significantly correlated to the histological grade and clinical stage of bladder cancer.Conclusion： The expression of Plk1 is closely related to the occurrence,progression of the bladder cancer,which may become new targets for cancer therapy.
