奥曲肽对人卵巢癌耐顺铂细胞株SKOV3/DDP移植瘤增殖的影响

摘　　要：目的探讨奥曲肽对人卵巢癌耐顺铂细胞株SKOV3/DDP裸鼠移植瘤生长的影响。方法将SKOV3/DDP细胞接种雌性BALB/c-nu/nu裸鼠右腋前皮下构建移植瘤动物模型,成瘤后随机分成4组。奥曲肽组(A):奥曲肽100μg.kg-1.d-1,用药28 d;顺铂组(B):顺铂4 mg.kg-1.d-1,成瘤后第1、8、15、22天用药;联合组(C):奥曲肽和顺铂联用;对照组(D):生理盐水对照。用药4周后处死裸鼠,取肿瘤标本计算瘤重和瘤体积,并采用免疫组织化学法检测瘤块中生长抑素受体2(SSTR2)和表皮生长因子受体(EGFR)的表达。结果各用药组肿瘤标本的瘤重、瘤体积均低于D组,C组的瘤重、瘤体积低于A、B组(P<0.01)。各组移植瘤细胞均有SSTR2表达;EGFR表达量D组显著高于其他各组(P<0.01),C组及A组EGFR表达量低于其他两组(P<0.01)。结论奥曲肽能抑制人卵巢癌耐顺铂细胞株SKOV3/DDP的裸鼠移植瘤的生长,并与顺铂有协同作用。Objective To study the influence of octreotide on the growth of human ovarian cancer SKOV3/DDP xenografts in BALB/c-nu/nu mice.Methods The xenografts model of nude mice was established by subcutaneously transplanting human ovarian cancer cisplatin-resistaent SKOV3/DDP cells.The mice were randomized to 4 groups with 5 mice each.The mice of group A were given octreotide 100 μg·kg-1·d-1 for 28 days,those of group B were given cisplatin 4 mg·kg-1·d-1 on the 1st,8th,15th and 22nd day after xenografts formed,and those of group C were given octreotide combined with cisplatin as in groups of A and B.The mice in group D were given normal saline as the controls.The mice were sacrificed on the 29th day for the measurements of size and weight of the xenografts.A routine observation of pathological section and immunohistostaining was conducted.Epithelial growth factor receptor（EGFR） and somatostatin receptor 2（SSTR2） were detected by immunohistochemistry.Results The weight and size were higher in group D than those in groups of A,B and C（P0.01）,which were lower in group C than those in groups of A and B（P0.01）.There were SSTR2 expressions in xenografts of 4 groups.Expression of EGFR was higher in group D than that in groups of A,B and C（P0.01）,which was lower in groups of C and A than that in groups of B and D（P0.01）.Conclusion Octreotide can significantly inhibit the growth of human ovarian cancer SKOV3/DDP xenografts in BALB/c-nu/nu mice and has a synergetic effect with cisplatin.
