摘  要:目的探讨威灵仙多糖对人乳腺癌MCF-7细胞凋亡的影响及其分子机制。方法不同浓度威灵仙多糖作用于MCF-7细胞48h后,荧光显微镜观察人乳腺癌细胞MCF-7细胞凋亡情况,RT-PCR法检测Bcl-2、Fas基因表达量的变化。结果威灵仙多糖能诱导MCF-7细胞凋亡,40mg/L时凋亡率为17.63%,实验组与对照组比较有显著性差异;并且上调Fas和降低Bcl-2表达。结论威灵仙多糖能抑制MCF-7细胞增殖,促进其细胞凋亡,其机制可能与激活Fas、抑制Bcl-2的基因表达有关。Objective:To investigate the influence of Radix Clematidis polysaccharide(RCP)on apoptosis of MCF-7 cells and reveal the mechanism.Methods:MCF-7 cells were treated with various of RCP,we observed cell apoptosis of MCF-7 cells by fluorescence microscope.We detected the levels of Bcl-2 and Fas gene expression in all groups by RT-PCR.Results:RCP has obviously apoptosis-inducing effects on MCF-7 cells.And apoptotic rate was 17.63%when the density of RCP was 40mg/L.It was found that there was a significant difference between the experiment groups and control group.It could effectively decrease the level of Bcl-2 gene expression and recrease the level of Fas gene expression.Conclusion:RCP could inhibit the growth and induce apoptosis of MCF-7 cells probably through down regulating Bcl-2 gene expression and up regulating Fas gene expression.
