摘 要:旨在研究MyoG基因内含子Ⅱ多态性与波尔山羊及其级进杂交后代部分生长性状的关系。以纯种波尔山羊及波尔山羊与唐山奶山羊级进杂交一代(F1)、二代(F2)和三代(F3)为研究对象,采用测序和PCR-RFLP方法对山羊MyoG基因遗传多态性进行分析,并研究基因型对初生体质量、1月龄体质量和断奶体质量的影响。结果在MyoG基因内含子Ⅱ的1 823bp处(依据序列号:FJ607135)发现了1个SNP(C→T),且存在3种基因型(AA、AB和BB),这3种基因型在波尔山羊和波唐级进杂交后代4个群体中的分布基本一致,B等位基因为优势等位基因。通过对有效等位基因数(Ne)、群体杂合度(H)、Shannons信息指数(I)和多态信息含量(PIC)等群体遗传参数的计算,发现此SNP是1个中、低多态位点。最小二乘分析结果表明,AA基因型与其它2种基因型比较有较小的初生体质量且达到显著水平(P<0.05),而对于1月龄体质量和断奶体质量,3种基因型间虽未达显著水平(P>0.05),但具AA基因型个体的表型值明显低于具其它2种基因型个体的表型值,AA、AB和BB 3种基因型在这3个生长性状的大小排列顺序均为BB>AB>AA。推测MyoG基因对山羊生长性状存在一定影响,提示选择带有B等位基因的个体有望提高山羊体质量相关的生长性状。To reveal the polymorphism of MyoG gene intron Ⅱ in Bore goat and its offspring and evaluate its relationship with the partial growth traits,single-nucleotide polymorphisms of MyoG gene intron Ⅱ were tested using PCR-RFLP and sequencing in the different goat populations including Bore goat and its offspring to Tangshan dairy goat,and the effects of the MyoG gene on the birth weight,the 1-month body weight and the weaning weight were also analyzed.The result of sequencing showed that one SNP(C→T) was found at 1 823 bp(Accession number: FJ607135) of MyoG gene intron Ⅱ,which existed three genotypes(AA,AB,BB).The distributions of 3 genotypes were basically identical in four goat populations,and allele B was the dominant.This SNP was a middle-low polymorphic locus by statistics of effective number of alleles(Ne),heterozygosity(H),information index(I) and polymorphism information content(PIC).Using least square analysis,it was showed that individuals with the AA genotype had significantly less(P〈0.05) birth weight than those with the BB and AB genotypes.Although there was no significant difference(P〉0.05) for the 1-month body weight and the weaning weight among the different genotypes,the phenotypic values of the individuals with AA genotype were obviously less than those with BB and AB genotypes,with the order of BB〉AB〉AA in three traits.These results suggest that the MyoG gene has some effects on the growth traits of goat,and the selection of B allele is favored with regard to the growth traits of goat.
