摘  要:目的探讨血小板反应素-1(TSP-1)在前列腺癌患者癌组织及其外周血中的表达及其意义。方法应用免疫组化技术检测TSP-1在前列腺癌组织(前列腺癌组)中的表达;以半定量RT-PCR技术检测TSP-1mRNA在患者外周血中的表达,并与前列腺增生患者(前列腺增生组)进行比较。结果前列腺癌组TSP-1阴性表达6例,弱阳性表达16例,阳性表达4例,增生组TSP-1弱阳性表达12例,阳性表达16例,无阴性表达,前列腺癌组TSP-1表达强度较增生组显著降低(P<0.01);TSP-1mRNA在前列腺癌组织中较前列腺增生组织中表达明显下调(P<0.01)。TSP-1mRNA在前列腺癌组外周血中的表达较增生组明显下调(P<0.01)。结论 TSP-1基因及蛋白表达水平在前列腺癌组织中明显下调,在前列腺癌患者外周血中表达水平明显下调。Objective To investigate the relationship between thrombospodin-1 expression and the development of human prostate cancer and its clinical significance. Methods The expression of TSP-1 in 26 cases of prostate cancer tissues and 28 hyperplastic prostate tissues, and the expression of TSP-1 mRNA in the peripheral blood of patients with prostate cancers were detected with immunohistochemistry methods and semi-quantitive RT-PCR. The relationship between the expression of TSP-1 and the clinic pathological features of prostate cancer was analyzed with statistical method. Results 6 cases of prostate cancer tissues showed no TSP-1 expression, 16 weakly positive and 4 positive. While 12 cases of hyperplastic tissues showed weakly positive and 16 positive. There was a significant correlation between the two groups. The expression of TSP-1 mRNA in the peripheral blood and prostate cancer tissues were down-regulated (P〈0.01) compared with that in the hyperplastic prostate tissues. Oonclusiorts The expression of TSP-1 was down-regulated in prostate cancer tissues, and the expression of TSP-1 mRNA was down-regulated in the peripheral blood of patients with prostate cancer.
