摘 要:目的:检测精神分裂症模型小鼠脑内c-Fos与尼克酰胺腺啶呤二核苷酸磷酸黄递酶(NADPH-diapho-rase,NADPH-d)的共存,探讨一氧化氮(nitricoxide,NO)与精神分裂症的关系,为深入研究精神分裂症的发病机制提供形态学和神经化学依据。方法:雄性昆明小鼠,随机分为生理盐水组和苯环己哌啶(phencyclidine,PCP)组,分别用自主活动观察、Sams-Dodd的刻板行为评分标准对模型小鼠的行为改变进行检测,同时用NADPH-d组织化学和c-Fos免疫组织化学双染法检测各组小鼠脑内c-Fos/NADPH-d阳性神经元的分布。结果:(1)PCP组小鼠出现明显的自发活动的增加(P<0.01)及显著的刻板行为(P<0.01);(2)PCP组小鼠脑内c-Fos与NADPH-d阳性神经元单纯表达的数量明显高于对照组,并且在伏隔核、屏状核及被盖背外侧核出现较多c-Fos/NADPH-d双标阳性神经元,生理盐水组小鼠脑内未见c-Fos/NADPH-d双标阳性神经元。结论:NO可能在谷氨酸NMDA受体功能异常引起的精神分裂症的病理过程发挥一定作用。Objective: To investigate the relationship between nitricoxide NO and the schizophrenia by detecting the expressions of c-Fos/ NADPH-d in the mouse brain,and to provide the evidence of morphology and neurochemistry for nosopoietic mechanism of schizophrenia.Methods: Male Kunming mice were randomized into two groups: normal saline group and phencyclidine(PCP) group.Their ethological changes were determined by spontaneous activity observation and Sams-Dodd stereotyped behavior scores respectively.Meanwhile,labeling with histochemistry-immunohistochemistry was performed to observe c-Fos/NADPH-d positive neurons of each group.Results:(1)Compared with the normal saline group,the PCP group mice had more spontaneous activities(P〈0.01),and showed more significant stereotype behavior(P〈0.01);(2)The number of simple c-Fos or NADPH-d single labeled neurons in the PCP group was significantly higher than that in normal saline group.More c-Fos/NADPH-d double-labeled neurons in the Acb(accumbens nucleus),Cl(claustrum) and LDTg(laterodorsal tegmental nucleus) were observed in the PCP group,while negative in the normal saline group.Conclusion: NO may play a role in the pathological process of schizophrenia caused by abnormal glutamate NMDA receptor function.
