摘  要:目的探讨环氧合酶(cyclooxygenase-2,COX-2)表达上调与人卵巢癌新生血管生成的关系。方法采用免疫组织化学法对80例卵巢癌组织中的COX-2进行检测;同时使用免疫组织化学法研究卵巢癌组织中VEGF及bFGF的表达情况,使用Weidner分析方法计算CD34标记的微血管密度(microvessel density,MVD)。结果 COX-2、VEGF和bFGF在卵巢癌中的表达分别为85.0%(68/80)、78.7%(63/80)和75.0%(60/80);VEGF和bFGF在COX-2阳性组表达率分别为92.6%(63/68)和88.2%(60/68),而在COX-2阴性组不表达;VEGF和bFGF在COX-2阳性组的免疫染色积分分别为17.21±2.81和16.21±2.43,COX-2与VEGF和bFGF有明显相关性(P<0.01)。MVD在COX-2阳性组和COX-2阴性组分别为54.21±16.61和36.21±10.21,两者有显著性差异(P<0.01),COX-2与MVD有明显相关性(P<0.01)。结论 COX-2表达上调与卵巢癌血管生成有密切相关,可能促进卵巢癌血管生成,共同促进卵巢癌的浸润转移;联合检测COX-2、VEGF和bFGF可能发展为卵巢癌有价值的预后指标。Objective To explore the correlations between up-regulation of COX-2 expression and angiogenesis in ovarian carcinoma.Methods The immunohistochemical expression of COX-2 were examined in eighty cases of ovarian carcinoma,and its contributions to the expression of VEGF and bFGF.Weidner analysis system was used to assess the MVD as a percentage of the endothelial area,and the MVD was assessed using CD34 immunohistochemistry.Results The expression rates of COX-2,VEGF and bFGF were 85.0%(68/80)、78.7%(63/80) and 75.0%(60/80),respectively.The expression rates of VEGF and bFGF were 92.6%(63/68) and 88.2%(60/68) in positive group of COX-2,and VEGF and bFGF were not expressed in negative group of COX-2.Immunohistochemical staining scores of VEGF and bFGF were 17.21±2.81 and 16.21±2.43 respectively in positive group of COX-2 and zero in negative group of COX-2.COX-2 was significantly correlated with VEGF and bFGF.MVD was 54.21±16.61 and 36.21±10.21 in positive and negative group of COX-2 respectively,and COX-2 was significantly correlated with ovarian carcinoma MVD(P0.01).Conclusions Up-regulation of COX-2 expression correlations with tumor angiogenesis in ovarian carcinoma,and it plays an important role in tumor genesis and development of ovarian carcinoma.Synergistic exploring of COX-2,VEGF and bFGF is a useful progress.
