摘 要:目的:探讨LIVIN和BAG-1蛋白在弥漫大B细胞淋巴瘤组织中的表达及其临床意义。方法:选取皖南医学院附属弋矶山医院2008年7月~2010年11月初治的弥漫大B细胞淋巴瘤组织标本31例,反应性淋巴结增生21例作为对照组,采用免疫组化S-P法检测LIVIN与BAG-1的表达,探讨其与弥漫大B细胞淋巴瘤的分期及预后的关系。结果:LIVIN在弥漫大B细胞淋巴瘤组织中表达率为64.52%,对照组表达率为23.81%(P<0.05);BAG-1在弥漫大B细胞淋巴瘤组织中表达率为61.29%,对照组表达率为28.57%(P<0.05)。LIVINⅠ期+Ⅱ期弥漫大B细胞淋巴瘤患者组织中表达率为50.00%,Ⅲ期+Ⅳ期弥漫大B细胞淋巴瘤患者组织中表达率为66.67%(P>0.05);BAG-1Ⅰ期+Ⅱ期弥漫大B细胞淋巴瘤患者组织中表达率为50.00%,Ⅲ期+Ⅳ期弥漫大B细胞淋巴瘤患者组织中表达率为62.96%(P>0.05)。LIVIN在弥漫大B细胞淋巴瘤生发中心亚型表达率为33.33%,非生发中心亚型表达率为77.27%(P<0.05);BAG-1在弥漫大B细胞淋巴瘤生发中心亚型表达率为22.22%,在非生发中心亚型表达率为77.27%(P<0.05)。结论:LIVIN和BAG-1在弥漫大B细胞淋巴瘤中高表达,在弥漫大B细胞淋巴瘤非生发中心亚型高表达。LIVIN和BAG-1的表达与弥漫大B细胞淋巴瘤的临床分期无关,与弥漫大B细胞淋巴瘤病理亚型有关。LIVIN和BAG-1可作为判断弥漫大B细胞淋巴瘤预后的生物学指标。Objective:To investigate the Livin and BAG-1 gene expression and its clinical significance in diffuse large B-cell lymphomas.Methods: The specimens were collected from 31 untreated cases of diffuse large B-cell lymphoma(DLBCL)from Yijishang Hospital of Wannan Medical Colledge between Jun.2008 and Nov.2010 and 21 cases of benign lymph nodes hyperplasia(BLH) were included as controls.Immunohistochemical staining(streptavidin-perosidase,SP) was used to detect the expressions of Livin and BAG-1 for estimation of the relationship between DLBCL and prognosis.Results: Livin gene was positively expressed in 64.52% of the DLBCL and in 23.81% of the controls(P0.05).Positive BAG-1 was 61.29% in DLBCL versus 28.57% in the benign lymph nodes hyperplasia tissues(P0.05).BAG-1 was expressed in 50.00% of DLBCL in stage Ⅰ-Ⅱ and 62.96% in stage Ⅲ-Ⅳ(P0.05).In germinal center B-cell-like(GCB)subtype of DLBCL,the expression of Livin was 33.33% versus 22.22% for BAG-1,and yet the two genes were equally expressed in non-GCB subtypes(77.27%)(P0.05).Conclusion: Overexpression of Livin and BAG-1 in DLBCL and non-GCB subtypes suggests that the two genes has no association with clinical stage but are related to subtypes.Clinically,Livin and BAG-1 can be an biomarker to estimate the prognosis of this disease.
