摘　　要：目的研究促凋亡基因PDCD5和Caspase-3 mRNA在多发性骨髓瘤(MM)中的表达,并讨论其在MM的发生、发展及预后中的意义。方法从收集40例MM患者及10例正常对照的骨髓标本中取骨髓单个核细胞(BM-MCs),采用实时荧光定量PCR(Real ti me PCR)的方法检测BMMCs中PDCD5和Caspase-3 mRNA的表达,同时收取血浆以检测β2微球蛋白(β2-MG)分泌水平,Durie-Sal mon(D-S)分期标准对MM患者进行临床分期。结果与对照组比较,PDCD5和Caspase-3 mRNA在MM组中表达明显减低(P<0.05),差异有统计学意义;复发/难治组患者PDCD5和Caspase-3 mRNA的表达仍保持较低水平;初发组与复发/难治组之间表达差异无统计学意义。PD-CD5和Caspase-3 mRNA表达与患者病程D-S分期和β2-MG(≥或<4.0 g/L)水平有负相关性(r=-0.86,P<0.05)。结论MM患者PDCD5和Caspase-3 mRNA表达水平降低,提示PDCD5和Caspase-3可能参与MM的发生发展,可能作为评判MM患者预后的新指标。Objective This study was aimed to explore the expression significance of apoptosis related gene PDCD5 and Caspase-3mRNA in MM.Methods Real-time polymerase chain reaction（Real time PCR） was used to detect mRNA of PDCD5 and Caspase-3 in 40 cases of MM.Results The expression of PDCD5 and Caspase-3 in patients with newly diagnosed and relapsed/refractory multiple myeloma was much lower than that in normal control and in patients reached plateau stage（P0.05）.There was no statistically significant difference between newly diagnosed and relapsed/refractory patients（P0.05）.The expressions of PDCD5 and Caspase-3 mRNA were related to stage of disease and β2-MG level（r=-0.86,P0.05）.Conclusion It is concluded that the expression of PDCD5 and Caspase-3 in patients with MM is low,which maybe an important factor for the pathogenesis of MM.PDCD5 and Caspase-3 can be as the indicators for predicting prognosis of MM.
