摘 要:目的:为了初步鉴定从土壤中分离的一株拮抗性能强且抗性稳定的菌株,并探索其抗菌物质的最佳发酵条件。方法:对拮抗菌一般特征采用苯酚品红染色法,芽孢采用孔雀石绿染色法,鞭毛采用西萨-基尔(Ceseres-Gill)染色法,生理生化测试采用常规的细菌生理生化测试方法进行鉴定。发酵条件分别开展培养基、发酵pH值、温度、时间的筛选。结果:经初步鉴定拮抗细菌B-05属枯草芽孢杆菌。抗菌物质最佳发酵条件为:在pH7.0的PDA培养基中,25℃恒温培养96h后发酵液的抑菌效果最佳。结论:发酵条件及抗菌性研究为抑菌物质的工厂化生产和应用提供了有价值的参考依据。Objective: The aim of this study was to identify initially a strain with strong antagonistic action and stable resistance, which was separated from the soil and to explore the best optimum fermentation conditions of its antibacterial substance. Methods: For the general characteristics of antagonistic bacteria, carbol fuchsia dyeing method was used. As for a geImna, schaeffer-fulton dyeing method was adopted. Ceseres-Gill dyeing method was introduced to the flagellum and the normal bacterial physiological and biochemical test methods was for physiological and biochemical test. The optimum fermentation conditions comprised developing medium and the sifting of pH value, temperature and time. Results: Mter the initial identification, antagonistic bacteria B-05 was included by Bacillussubtilis. The best optimum fermentation conditions for the antimicrobial substances was kept constant temperature of 25 ℃ for 96 h in the pH 7.0 PDA medium. Conclusion: The optimum fermentation conditions and the antimicrobial study offered valuable references for the production and application of antibacterial substances.
