摘 要:目的探讨ATP结合盒(ABC)转运体ABCG2,ABCB5在皮肤黑素瘤中的共表达及意义。方法采用免疫组化单/双重染色法检测30例皮肤黑素瘤、30例皮内痣及5例A375细胞移植瘤中ABCG2,AB-CB5的表达与共表达。结果①ABCG2在黑素瘤、皮内痣中阳性率分别为63.33%和0,两者相比差异有统计学意义(P<0.05),阳性细胞率平均值分别为(25.50±9.47)%和0,前者明显高于后者,在A375细胞移植瘤中阳性率和阳性细胞率平均值分别为100.00%和(27.00±8.04)%;②ABCB5在黑素瘤、皮内痣中阳性率分别为73.33%和0,两者相比差异有统计学意义(P<0.05),阳性细胞率平均值分别为(28.75±2.22)%和0,前者明显高于后者,在A375细胞移植瘤中阳性率和阳性细胞率平均值分别为100.00%和(30.50±4.66)%;③ABCG2和ABCB5在黑素瘤、皮内痣中共表达阳性率分别为60.00%和0,两者相比差异有统计学意义(P<0.05),共表达阳性细胞率平均值分别为(23.00±2.94)%和0,前者明显高于后者,在A375细胞移植瘤中阳性率和阳性细胞率平均值分别为100.00%和(24.50±4.04)%。结论皮肤黑素瘤与A375细胞移植瘤中存在ABCG2,ABCB5的阳性表达和共表达,提示皮肤黑素瘤的多药耐药可能与ABCG2和ABCB5有关。Objective To study co-expression of ABC transporter ABCG2 and ABCB5 in cutaneous malignant melanoma(CMM). Methods The expressions and co-expressions of ABCG2 and ABCB5 in 30 CMM, 30 intradermal nevus and 5 transplanted tumors of A375 cells in nude mice were detected by immunohistochemical technique. Results (1)The positive rates of ABCG2 expression in CMM and intradermal nevus were 63.33% and 0. There were significant statistically differences between them(P 〈 0.05 ). The mean values of positive cell rates were (25.50 ± 9.47 ) % and 0. There were significant differences between them. In A375 cells transplanted tumor the positive rates and the mean values of positive cell rates were 100.00%, (27.00 ± 8.04) %. (2)The positive rates of ABCB5 expression in CMM and intradermal nevus were 73.33% and 0. There were significant statistically differences between them ( P 〈 0.05 ). The mean values of positive cell rates were (28.75 ± 2.22) % and 0. There were significant differences between them. In A375 cells transplanted tumor the positive rates and the mean values of positive cell rates were 100.00%, (30. 50 ± 4.66) %. (3)The positive rates of ABCG2 and ABCB5 co-expression in CMM and intradermal nevus were 60.00% and 0. There was significant statistically differences between them( P 〈 0.05 ). The mean values of positive cell rates were (23.00 ± 2.94) % and 0. There were significant differences between them. In A375 ceils transplanted tumor the positive rates and the mean values of positive cell rates were 100.00%, (24.50 ± 4.04)%. Conclusions There were expressions and co-expressions of ABCG2 and ABCB5 in CMM and A375 cells transplanted tumor, these results imply that ABCG2 and ABCB5 maybe associated with the multidrug resistance in the chemical therapy of CMM.
