吲达帕胺对脂多糖诱导新生大鼠心肌细胞损伤后TNF-α表达的影响

摘　　要：目的探讨吲达帕胺对脂多糖(LPS)诱导乳鼠心肌损伤后TNF-α表达的影响。方法取SD新生大鼠(1～3 d)45只,分离心肌细胞培养,在培养的第4天按随机数字表法分为3组:正常对照组(对照组,行常规培养)、LPS组(加入LPS 1 mg.L-1,处理6 h)、吲达帕胺+LPS组(吲达帕胺组;加入吲达帕胺0.01 mg-1、处理12 h,LPS 1 mg-1、处理6 h)),每组15只。分别采用MTT、RT-PCR、Western Blotting法检测心肌细胞存活率、TNF-αmRNA、TNF-α蛋白的表达。结果细胞存活率:LPS组较对照组明显减少(P<0.01);吲达帕胺组较LPS组明显增高(P<0.01),与对照组相比差异无统计学意义(P>0.05)。TNF-αmRNA的表达:LPS组较对照组明显增加(P<0.01);吲达帕胺组较LPS组明显降低(P<0.01),而较对照组差异无统计学意义(P>0.05)。TNF-α蛋白的表达:LPS组较对照组明显增加(P<0.01);吲达帕胺组较LPS组明显减少(P<0.01),而较对照组差异无统计学意义(P>0.05)。结论吲达帕胺可下调LPS诱导的心肌细胞损伤后TNF-α的表达,减少心肌损伤。Objective To investigate the effect of indapamide on TNF-α expression in neonatal rat cardiocyte injury induced by lipopolysaccharide（LPS）.Methods Cultured cardiocytes from 45 neonatal SD rats aged 1～3 days were randomly divided into three groups on the 4th culture day：control group,LPS group and indapamide＋LPS group,with 15 rats in each group.LPS group was treated with LPS（1 mg·L-1） for 6 hours.Indapamide＋LPS group was treated with indapamide（0.01 mg·L-1） for 12 hours followed by LPS（1 mg·L-1） for 6 hours.Cardiocyte viability,TNF-α mRNA expression and TNF-α protein expression were measured by MTT,RT-PCR and western blotting,respectively.Results Cardiocyte viability in LPS group was significantly lower than that in control group and indapamide＋LPS group（P0.01）,but no differences were observed between indapamide＋LPS group and control group（P0.05）.The expression of TNF-α mRNA and protein in LPS group was obviously higher than that in control group and indapamide＋LPS group（P0.01）,but no differences existed between indapamide＋LPS group and control group（P0.05）.Conclusion Indapamide can dowz-regulate the expression of TNF-α to alleviate cardiac damage induced by LPS.
