摘　　要：[目的]探讨COX-2抑制剂对白血病细胞株U937中COX-2及VEGF表达的影响。[方法]培养U937细胞株,每次实验取12瓶5mlU937细胞液,分成3组,加入COX-2抑制剂试剂,使每组药物终浓度分别为5.0、10.0和50.0μmol/L。将3组细胞分别孵育6、12、24h后,观察不同药物终浓度组的细胞形态变化。采用流式细胞术检测COX-2和VEGF蛋白表达情况。[结果]COX-2抑制剂可抑制COX-2蛋白表达,并呈时间剂量依赖性。COX-2抑制剂作用后,U937细胞凋亡率增加,增殖受抑。[结论]COX-2抑制剂可抑制VEGF、COX-2表达,有希望用于治疗恶性血液病。[Purposel To investigate the influence of COX-2 inhibitor on the expression of COX-2 and VEGF in U937 cells. [Methods] U937 cells were cultured, 12 bottles of U937 cells were divided into 3 groups in every test, final concentration of COX-2 inhibitor was 5.0, 10.0 and 50.0μmol/L, respectively. The morphological changes of different concentration group were observed after incubated for 6h, 12h and 24h, respectively. COX-2 and VEGF protein expression were detected by flow cytometry. [Results] The expression of COX-2 and VEGF protein was inhibited by COX-2 inhibitor, with time/dose-dependent manner. The apoptosis of U937 cells was induced and the growth was inhibited by COX-2 inhibitor, [Conclusion] COX-2 inhibitor can inhibit COX-2 and VEGF expression, it might be hopeful for the treatment for hematologic malignancies.
