外周血血浆和白细胞EBVDNA检测的比较

摘　　要：目的比较荧光定量PCR检测外周血血浆和白细胞EB病毒DNA的结果,探讨荧光定量PCR检测血浆和白细胞EB病毒DNA的一致性。方法收集2009年3月至2011年3月疑似EB病毒感染患者的外周血(EDTA-K3抗凝)814例,其中鼻咽癌患者18例,非鼻咽癌的其他患者796例;采用实时荧光定量PCR技术同时同步检测血浆和白细胞的EB病毒DNA含量,比较鼻咽癌患者及其他非鼻咽癌患者的外周血血浆和白细胞EB病毒DNA的检测结果。结果 18例鼻咽癌患者外周血血浆和白细胞检测EB病毒DNA的阳性率相同,均为61%(11/18);其EB病毒DNA含量水平也没有统计学差异(P>0.05)。796例其他非鼻咽癌患者血浆和白细胞检测EB病毒DNA的阳性率分别为1.2%(10/796)和7.4%(60/796),两者在统计学上有显著差异(P<0.05)。结论荧光定量PCR检测鼻咽癌患者外周血血浆和白细胞EB病毒DNA的阳性率及其含量没有差异性(P>0.05);而检测非鼻咽癌患者白细胞EB病毒DNA的阳性率高于血浆。Objective To compare levels of EB virus DNA in plasma and leucocytes detected by Fluorescence Quantitative PCR,and disscuss the consistency between results of EB virus DNA in plasma and leucocytes detected by PCR.Methods Peripheral blood samples（EDTA-K3 Anticoagulanting） of 814 patients were collected,which were suspected by EB virus.Among 814 patients,18 patients suffered from nasopharyngeal carcinoma,while other 796 persons were diagnozed other non-nasopharyngeal carcinoma.Contents of EB virus DNA in plasma and leucocytes were detected simultaneously with the real-time fluorescence quantitative PCR.Results For 18 nasopharyngeal carcinoma patients,positive rates of EB virus DNA in plasma and leucocytes were 61%（11/18）;contents of EB virus DNA in plasma and leucocytes were not statistically different（P 0.05）.For 796 non-nasopharyngeal carcinoma patients,positive rates of EBV DNA in plasma and leucocytes were 1.2%（10/796） and 7.4%（60/796） respectively,with statistically significant difference（P 0.05）.Conclusion There is no statistical difference in levels of EB virus DNA in both plasma and leucocytes of nasopharyngeal carcinoma detected with fluorescence quantitative PCR.For patients of non-nasopharyngeal carcinoma,the positive rate of leucocytes is higher than that of plasma.
