摘 要:目的观察大鼠痫性发作后各时间点血清、脑脊液中降钙素基因相关肽(calcitonin gene related peptide,CGRP)含量的变化及其在海马组织的表达,以探讨CGRP与癫痫的关系。方法雄性SD大鼠48只随机分成动物模型组(40只)和对照组(8只),采用侧脑室注射红藻氨酸(kainic acid,KA)建立癫痫动物模型。在痫性发作后6h、12h、24h、72h、1周抽取血液和脑脊液,用ELISA方法测定血清和脑脊液中CGRP含量,用Nissl染色法观察海马病理改变,用免疫组织化学的方法观察各组大鼠海马CGRP蛋白的表达情况。结果血清和脑脊液中CGRP含量在痫性发作后6 h逐渐降低,在24 h最低。KA致癫痫后24h、72h及1周,大鼠海马CA3区和齿状回CGRP蛋白大量表达,伴随大量神经元丧失、尼氏体减少。结论 CGRP与癫痫关系密切,血清和脑脊液CGRP水平可用作反映癫痫所致脑损伤的早期生化指标。【Objective】 To investigate the levels of calcitonin gene related peptide(CGRP) in serum and cerebrospinal fluid(CSF) and the expressions of CGRP in hippocampal tissues of epileptic rats,in order to study the relationship between CGRP and epilepsy.【Methods】 48 male SD rats were randomly divided into model group(n =40) and control group(n =8).Animal models of epilepsy were established in SD rats by intracerebroventricular injection of kainic acid(KA).The levels of CGRP in serum and CSF were measured by ELISA at 5 different time points(6 h,12 h,24 h,72 h and 1 w after KA-induced seizures).The pathologic changes of the hippocampal tissues were studied by Nissl staining and the expressions of CGRP was observed by immunohistochemical staining.【Results】 The levels of CGRP in serum and CSF gradually decreased 6 h after seizures and reached the valley at 24 h.At 24 h,72 h and 1 w after seizures,the numbers of CGRP-immunopositive neurons in hippocampus CA3 area and dentate gyrus increased significantly with abundant neurons loss and Nissl bodies reduction.【Conclusion】 A compacted relation exists between CGRP and epilepsy,the level of CGRP in serum and CSF may be used as an early indicator in diagnosis of brain injury in epileptic rats.
