摘  要:目的观察小分子干扰RNA(siRNA)沉默livin基因在食管癌细胞中的表达情况,探讨livin基因对食管癌细胞生长、凋亡的影响。方法自行设计两条针对livin基因的siRNA:livin—sh-1,livin—sh-2,构建相应的表达载体分别转染至对数生长期的食管癌,经G418筛选后采用逆转录聚合酶链反应(RT—PCR)、MTT、TUNEL分别比较检测转染前后食管癌细胞livin基因的表达。结果siRNA对照组与空siRNA载体组的livinw/β mRNA表达差异无统计学意义(P〉0.05);但转染siRNA组Livinl/2mRNA表达显著降低,明显低于siRNA对照组和空siRNA载体组(P〈0.05)。细胞生长曲线测定经F检验三组间差异无统计学意义。转染siRNA载体沉默livina/β表达,细胞凋亡率显著增加。结论siRNA沉默livin基因能抑制食管癌细胞的生长,促进食管癌细胞的凋亡,livin基因有可能成为食管癌治疗的新靶点。Objective To investigate the feasibility of small interfering RNA(siRNA) for inhibi- ting livin gene expression, and the effect of livin gene on proliferation and apoptosis of esophageal cancer cells. Methods Two siRNA ( livin - sh - 1 and livin - sh - 2) that specifically target that the livin gene were designed based on their coding regions and were synthesized in vitro. To construct eukaryotic expres- sion vector pIRES -AcGFP -livin, they were then transfected into esophageal cancer cells and G418 was adopted to select positive clones. Expression level of livin mRNA was detected by reverse transcription poly- merase chain reation (RT -PCR), MTI' and TUNEL. Results Compared with cells without siRNA transfec-tion, siRNA control group in livinoα/β decreased significantly (P 〈 0. 05 ) ; while those of cells in siRNA group increased significantly (P 〉 0. 05).Cell growth curve of all the three groups by can make the Livinα/β expression in silence Conclusions siRNA can silence livin gene analysis of variance had no significance difference, siRNA group growth slowed and it's apoptosis rate was increased significantly. expression in esophageal cancer cells.
