摘 要:目的:探讨电针对海洛因成瘾大鼠学习记忆及海马神经细胞凋亡的影响及机制。方法:采用递增法建立海洛因成瘾大鼠模型,设对照组、成瘾组、成瘾+电针处理组(电针组),三组大鼠跳台实验测试大鼠学习记忆,并取三组大鼠脑海马组织,运用流式细胞仪检测细胞凋亡)。结果:跳台实验结果表明:与对照组比较,成瘾组大鼠学习记忆成绩降低(P〈0.01),电针组大鼠学习记忆成绩有所改善(P〈0.01);流式细胞仪结果显示:与对照组相比,成瘾组大鼠海马细胞凋亡率增加(P〈0.01),电针组大鼠海马神经细胞凋亡率较成瘾组降低(P〈0.05)。结论:海洛因成瘾大鼠海马神经细胞凋亡增加;电针针刺治疗可抑制海洛因引起的海马神经细胞凋亡;海洛因引起海马神经细胞凋亡可能是海洛因损害学习记忆的机制之一,电针针刺治疗可改善海洛因成瘾大鼠的学习记忆,其机制可能与针刺可抑制海马神经细胞凋亡有关。Objective.. To observe the effect of acupuncture on the learning,memory and apoptosis of hippocampal neurons in heroin-addict rats and investigate its potential mechanism. Methods: Thirty Wis ter rats were randomly divided into the control group, the heroin-addict group and the acupuncture group. Three groups were respectively conducted in step clown test, which was usecl to detect the learning and memory of rats. Apoptosis of neurocytes in rats hippocampus were measured by means of FCM. Results:Step-down-test results showed that the dependence of learning and memory in heroin addict rats was reduced (P〈0.01), electro acupuncture improved learning and memory in rats (P〈0.01); Flow eytometry showed that apoptosis rate of neurocytes in rats hippoeampus in the heroin addict group was significantly higher than that of the control group(P〈0. 01 ), while compared with the heroin-addict group, the apoptosis rate of neurocytes in the acupuncture group obviously more decreased(P〈0.05). Conclusion: Hippocampal neuronal apoptosis increases in the heroin-addict group while apoptosis rate in the acupuncture group decreases. One possible mechanism that heroin may damage the learning and memory is presumed that heroin induce apoptosis of hippocampus in rats. HANS acupuncture treatment can improve learning and memory of heroin-addict rats and its mechanism may be related to the acupuncture, which can inhibit the hippocampal neuronal apoptosis.
