摘 要:目的:研究低强度超声联合原卟啉IX对人舌鳞癌SAS细胞增殖抑制及诱导凋亡作用。方法:SAS细胞分成对照和实验组,台盼兰和四甲基偶氮唑盐(MTT)法检测细胞活性,流式细胞仪检测细胞凋亡,透射电镜观察超微结构改变。结果:连续波强度结合100g/L原卟啉IX作用60s对SAS细胞增殖有显著的抑制作用,呈时间依赖性。透射电镜观察到凋亡小体及线粒体等细胞器改变。结论:低强度超声激活原卟啉IX诱导SAS细胞凋亡的作用机制可能与线粒体凋亡途径相关。Objective: To study the low--intensity ultrasound combined with protoporphyrin IX on human tongue cancer cell line SAS proliferation and apoptosis. Methods: SAS cells were divided into control and experimental group, Cell activity was detected by trypan blue and tetrazolium salt (MTT), and cell apoptosis was analyzed by flow cytometry. Results: The proliferation of SAS cells was inhibited significantly by the continuous--wave strength Combined with 100ug/ml protoporphyrin IX for 60s. The growth inhibition effect was time--dependent. The chan- ges of apoptotic bodies and mitochondrion were observed by TEM. Conclusion: The mechanism that low intensity ultrasound induced apoptosis of SAS cells significantly by activating protoporphyrin IX may be associated with mito- chondria apoptotic pathway.
