摘 要:目的观察低渗环境对体外培养星形胶质细胞增殖和细胞周期的影响。方法采用原代培养的大鼠皮层星形胶质细胞,实验分为对照组和低渗组;低渗1h后恢复正常全培养基,在不同时间点(0、12、24、48h),应用流式细胞技术以及免疫细胞荧光三标法(B rdu/GFAP/DAPI)检测各实验组星形胶质细胞增殖及细胞周期分布的情况。结果我们发现低渗干预星形胶质细胞1h并恢复正常培养基后12、24、48h,对照组和低渗组的B rdu阳性率和细胞周期分布无显著性差异(P>0.05)。结论低渗干预1h对星形胶质细胞的增殖速率及细胞周期的进展无显著影响。Objective To observe the effect of hypotonic solution on primary astrocytes proliferation and cell cycle progression.Methods Primary cultured rat cortex astrocytes were randomly divided into two groups: control group and hypotonic group.In the present study,we applied hypotonic solution followed by recovery in the cultured rat astrocytes.At different time points(0,12,24,48h),the astrocytes proliferation rates were evaluated by Brdu incorporation assay,while the cell cycle analysis was performed by flow cytometry with prodium iodine(PI) staining.Results Compared with the control group,there were no statistical change of the astrocytes proliferation rates and cell cycle distributions after hypotonic treatment.Conclusion Hypotonic treatment couldn't induce proliferation and cell cycle progression of cortex astrocytes in vitro.
